For the last ten years, within molecular life sciences, the reproducibility crisis discourse has been embodied as a crisis of trust in scientific images. Beyond the contentious perception of “questionable research practices” associated with a digital turn in the production of images, this paper highlights the transformations of gel electrophoresis as a family of experimental techniques. Our aim is to analyze the evolving epistemic status of generated images and its connection with a crisis of trust in images within that (...) field.From the 1980s to the 2000s, we identify two key innovations (precast gels and gel docs) leading to a “two‐tiered” gel electrophoresis with different standardization procedures, different epistemic statuses of the produced images and different ways of generating (dis)trust in images. The first tier, exemplified by differential gel electrophoresis (DIGE), is characterized by specialized devices processing images as quantitative data. The second tier, exemplified by polyacrylamide gel electrophoresis (PAGE), is described as a routine technique making use of image as qualitative “virtual witnessing.” The difference between these two tiers is particularly apparent in the ways images are processed, even though both tiers involve image digitization. Our account thus highlights different views on reproducibility within the two tiers. Comparability of images is insisted upon in the first tier while traceability is expected in the second tier. It is striking that these differences occur not only within the same scientific field, but even within the same family of experimental techniques. In the second tier, digitization entails distrust, whereas it implies a collective sentiment of trust in the first tier. (shrink)

Background: Increasing resistance of Pseudomonas aeruginosa to ciprofloxacin in ICU/burn units has created a problem in the treatment of infections caused by this microorganism. -/- Methods: Fifty P. aeruginosa strains were isolated from burn patients hospitalized in the Kerman Hospital during May 1999-April 2000 and were tested for in-vitro sensitivity to different antibiotics by disc diffusion breakpoint assay. The isolates were subjected to minimum inhibitory concentration (MIC) test by agar dilution method. Existence of the plasmids was also investigated in the (...) isolates. -/- Results: Thirty-four patients infected with ciprofloxacin strains showed MIC of 8 mg/ml [p<0.001]. Sixteen patients were infected with sensitive strains exhibiting MIC range of 0.0125-0.125 ± 0.033 mg/ml. The isolates were also also resistant to other antibiotics [p<0.001]. Plasmid isolation and agarose gel electrophoresis (0.7%) revealed three plasmid bands in strains 8 and 16, and one band in strain 35. -/- Conclusion: The emergence of ciprofloxacin resistance of P. aeruginosa in burn patients is alarming since this antibiotic has only recently been introduced onto the market in Iran. One important observation was that some isolates exhibited cross resistance to other antibiotics. Furthermore, some strains were carriers of plasmids which might have acted as the potential source of acquired resistance in the hospital setting. -/- . (shrink)

Enric F. Gel has recently argued that classical theism enjoys a significant advantage over Graham Oppy's naturalism. According to Gel, classical theism – unlike Oppy's naturalism – satisfactorily answers two questions: first, how many first causes are there, and second, why is it that number rather than another? In this article, I reply to Gel's argument for classical theism's advantage over Oppy's naturalism. Hannah Arendt disavowed the title of “philosopher,” and is known above all as a political theorist. But the relationship between philosophy and politics animates her entire oeuvre. We find her addressing the topic in The Human Condition (1958), in Between Past and Future (a collection of essays written in the early 1960s), and in Men in Dark Times (another collection of essays, this one from the late sixties). It is treated in her Lectures on Kant’s Political Philosophy, composed during the (...) seventies, and also in the posthumous Life of the Mind, two of three projected volumes of which were complete when she died in 1975. Certainly, Arendt’s thought cannot be understood without taking into account her deep suspicion of and equally deep commitment to philosophy in the context of political reflection. For all that, her writings on this abiding preoccupation do not gel into a systematically articulated theory or programmatic statement. Instead, they reflect Arendt’s appreciation of what remained for her a “vital tension” – an enigma. (shrink)

ABSTRACT: A report on the pioneering of a new pedagogy designed to challenge students to use and improve their memory, increase their awareness of logical fallacies and tacitly embedded contradiction(s) and sensitize them to the deeply symbolic nature of thought in all its expressions (math, logos, music, picture and motor skills), as created, by the author, from in situ research at a senior level (ESL) course in Storytelling at one of East Asia’s premiere second languages university, and from teaching children (...) in several university summer programs. -/- As a Visiting Professor of Foreign Languages, I recently had a glorious opportunity to innovate curriculum and teaching techniques in an East Asian university classroom. Under the auspices of teaching senior level thematic courses in English as a Foreign Language, or more specifically, “storytelling,” I was blessed with the opportunity to innovate both on the level of curriculum and pedagogy. -/- But let me backtrack to set up my story from the start. All of this began in a children’s ESL class at a private language institute where I was given some newly formed intermediate level English classes for middle school students on a new campus that had yet to form a library. While waiting for my standard textbooks to arrive I began innovating with one of my own childrens’ stories titled The Tale of the Purple People Eater (a three page mythic story written in the late 90s. Please see appendix). In working with this story, because I envisioned presenting it orally (from memory) at first as a way to introduce students to memory arts exercises, I was compelled to read it over many times, which, to my surprise revealed a troubling feature: it was riddled with contradictions in every paragraph. At any rate, with the middle school aged students I only touched on this feature very briefly and focused more on language acquisition exercises. The tale nevertheless impressed upon me this bizarre aspect of language where a story can appear naturally cogent on the surface, but can also come apart at the seems if taken “literally.” Hence, later while teaching Storytelling in university with a rather dry textbook, I was invited to supplement the class with exercises of my own. Here I was curious to try work shopping the ‘Tale of The Purple People Eater’ with young adults. This involved three stages: 1) Memory work — or engaging with the unpublished story orally; 2) Exploring the contradictory nature of language and thought; 3) Probing the “deeply-symbolic” nature of meaning. -/- Of course, all of these levels have a long history of precedence in our civilization. Memory arts — though defunct now — had been highly developed in the ancient world, not least among bards who performed oral recitations from memory of the entire Illiad and Odyssey. Dialectics — now transmuted into the debate for show and prizes — had a deeper epistemological mission to discover truth and achieve enlightenment (i.e. Socrates, Boethius and St-Augustine). And if it could not be done via contradictory-prone rationality, it would do it by re-attaching or re-membering broken meanings via the sym-bol (“throwing together”), as per our lineage of platonic poets who created a world compelling enough to make us ask where Jesus went during his (underworld journey) three days in the crypt? Or, and, on what terrain did Dante travel? -/- Lets look at all three: Memory work can be very simple: it amounts to an oral recitation of the story in class followed by a homework assignment wherein the students are asked to reproduce the story (in writing) from memory. They can then compare ‘notes’ in class and by working in groups attempt to produce one version per group of the story in question. In this way we invite them to engage with ‘live’ stories as did our oral-history ancestors. Likewise, by writing it down (“for the first time”) they are re-living the act of our first bardic writer(s) who wove together a collection of competing versions of a popular legend to produce a standard. Very primal work! If Plato thought that writing meant the end of memory in his time, imagine what smart phones are doing to memory now. Contradictions too are easy to work with. It suffices to project a written version of the teacher’s story on the classroom screen — no note taking! — and let students have ample time to read it over and over, at first to check with their own memorized version, and later to seek out logical contradictions in the prose. Any way to ‘repair’ the story? Does it lose credibility because of contradictions? Can it be real without being true? Can it be true without being real? Do reality (L. res = matter) and truth (Gk. a-leithea = un-forgetting) etymologically speaking, share the same domain? -/- Further, if below the surface-meaning the story falls apart, its apparent mythical meaning immediately is called into question. This is a good time to ask the students to propose a (deeper) interpretation. On the surface, the obvious meaning of the Tale of the Purple People Eater, a world in which a purple ogre predatorily feasted on the purple race of the United Peoples of Benetton (a multi-racial nation representing every “skin colour of the rainbow”), revolves around racism, and indeed everyone save for one, thought the story was about racism. -/- I, the author, had the challenge of discovering the deeper meaning of my own story. Because it was written inspirationally one morning, the way we might record the memory of a dream from the night before in a journal, I also was ignorant of the story’s meaning. As luck would have it, a few days before I was to give my class feedback on their interpretations, a meditation on the color “purple” led to the book “The Color Purple” which I first heard of from a friend who, in my reply to a question about the book she was reading (in 1985), summarized it succinctly in one sentence about a black woman’s sexual awakening and liberation. I quickly got the book from the university library as well as the film which I viewed several times before my next class. -/- To my utter amazement, after reading the Tale of the Purple People Eater in light of the Color Purple and adjusting the meaning of color in the tale from the color of people’s complexion to the “color of people’s souls” (to commensurate with the film) as depicted in their clothing and other accoutrements (e.g. automobiles) as reflective of the “health of their souls” (white for revolt, purple for majesty, etc…) where the enslaved Celie is surrounded by drab lifeless colors in the beginning of the film in contrast to the liberated woman she has become in the end of the story symbolized by the extraordinary intensity of the purple shawl that bedecks her like an African queen, the tale can be considered, in part, as a kind of back story of the inner evolution of the book’s and film’s protagonist. -/- Many other cross-polenised metaphors come to the fore: Hippocrates in the tale is the mail man in the film; St-Yves anti-ageing gel is a metaphor for remaining in “childhood (sexual) slavery”; the Ex-Thrax vaccine is the spiritual laxative in the form of letters from Africa (from Ex-Lax and Thrace, a “wild place”) that eases Celie out of her yoke; the egg-shaped conference table with the pointy end in the tale is the kitchen table in the last meeting where Celie threatens her husband with the pointy end of the knife in the film… -/- Of course, the students could not be expected to decode the meaning of a story of this nature whose collective unconscious connotations the author himself had to go to great lengths to excavate almost like a detective of the subconscious. What was important here was the shared engagement between mind and narrative and how it acted as a vehicle for 1) re-membering, 2) becoming aware that all language is a metaphorical cover and 3) that the deep symbol and fused metaphor are divided not by external time and space but by external reality (in the Latin sense, res) and internal truth (in the Greek sense, aleithea). Myth then, in its highest meaning is a hybrid form (of these two) of narrative. Hence both parts real and true. (shrink)

It argues that the use and prevalence of video-sharing technologies on sexual social networking websites has a direct impact on notions of sexual identity. Though sometimes pitted against one another and at times contradictory, the ideas of Michel Foucault and Judith Butler on the nature and expression of sexuality and gender identities in fact gel rather well, and both can help us to gain a deeper and more (...) rounded picture of the impact and importance of the burgeoning phenomenon of internet dating websites in general, and sexual social networking in particular. Söz konusu sorunun üstesinden gel- mek, yerelliğin neliğini ortaya çıkarabilmek için, yerelliğe felsefi bir bilinçle yaklaşılarak, kavramın içeriğinin, kavram haritasının ve ilkelerinin belirginleştirilmesi gerekmektedir. Bu amaçla çalışmamızda ilkin yerellik arayışının modernleşmeyle birlikte sömürgecilik (...) ve Batılılaşma karşısında ulusçuluk, postmodernizm ve çoklu modernite görüşleri içeri- sinde nasıl tesis edilmeye çalışıldığı ve yetersizlikleri panoramik olarak sunulmuş, ardın- dan kavramın ne olmadığı üzerinde durulmuştur. Araştırma sonucunda yerelliğin üç temel ilkesi olduğu tespit edilmiştir: Yerellik bir insan-mekân sorunudur; salt bir yerellik bulunmamaktadır ve yerellik görecelidir; saf bir yerellik yerine yerellik, sentez ve yerelleşme süreci olarak anlaşılmalıdır. Söz konusu ilkelerden hareketle ve yerellik kavramının içeriğinin sorunla- rına karşı, yerellik yerine kültürel(lik) kavramının kullanılması önerilmiştir. The purpose of this article is to discuss the meaning and content of the concept of locality. Although locality is often referred to as a reflex of nationalization it has remained far from being evaluated in terms of philosophy. For this reason the concept of locality is equated with similar concepts and wrapped in an ideological content and its meaning becomes increasingly uncertain. In order to overcome the aforementioned problem and to reveal the nature of locality, and to clarify the content, concept map and principles of the concept it is necessary to approach locality with a philosophical consciousness. Firstly how the search for locality is tried to be established within the views of nationalism, postmoder- nism and multiple modernity against colonialism and Westernization with modernization and its inadequacies are presented panoramically, then it is emphasized what the concept is not. Localization, which is pointed out to be considered separately from localism and indigenousness, has been examined in terms of its relationship with people, space and history and attention has been drawn to localization. As a result of the research it has been determined that locality has three basic principles: Locality is a human-space problem; there is no mere locality and locality is relative; rather than a pure locality, locality should be understood as a process of synthesis and localization. Based on these principles and against the problems of the content of the concept of locality it is suggested to use the concept of cultural instead of locality. (shrink)

In electrochemical assisted abrasive flow machining higher abrasion of the material was detected due to the combine effect of ECM and AFF processes. In Electrochemical aided abrasive flow machining a electrolyte is added to the prepared media .This media consist a kind of polymeric carrier and abrasive particles that are hydrocarbon gel, (...) Al2O3, Silicon based polymer, and NaI Sodium iodide as electrolytic salt. In this experimental research different process parameters such as voltage, abrasive concentration, Number of cycle, molal concentration and diameter of rod were considered at different levels for response characteristic of surface roughness Ra and material removal MR based on Taguchi method using standard L27 orthogonal array OA for the plan of experimentation. In this work, polystyrene/waste tires composites were prepared using the melt-mixing method.. The effect of the preparation conditions of the PS/waste tires composite is investigated in this work by using Gel permeation chromatography (GPC) technique. Stability at elevated temperatures were found to increase with the addition of waste tires to PS as confirmed by the results. GPC results indicate that the preparation conditions of the PS/waste tires composite has no effect on the Ps chains as indicated by GPC technique. The (...) signals of the GPC for unfilled and filled PS are similar. That evidences no thermo-mechanical chain degradation is taken place during the addition of the waste tires under the conditions of the melt-mixing process . (shrink)

In contemporary discussions on identity, difference and subjectivity, the thinking of Judith Butler has played a significant role, viewing identity as a problematic question of how the subject is constituted by historical structures of power that deprives it of original ownness and substantiality. As this situation for Butler stems neither from a historical occurrence, nor is clearly ontological, the question of the origin of this problem of identity and power remains. Through the thinking of Martin Heidegger, the present study develops (...) the argument that this problem indicates the completion of subject metaphysics, not its overcoming. When identity is thought and lived in terms of subjectivity, power will be its condition. This argument is first developed through a comparison of Butler’s and Heidegger’s respective appropriations of He-gel’s philosophy, together with an independent account of Hegel’s thinking. For Hegel, identi-ty involves a strive for being as a mediation of subjectivity and objectivity, where conscious-ness incorporates both beyond their split. According to Butler, this is impossible, as subjectivi-ty and objectivity are multiple, temporal and shared. For Heidegger however, the problem lies in the very thought of being as a connection of subjectivity and objectivity, and the under-standing of the human as the ground of this reconciliation. I will thereafter examine Heidegger’s “The Principle of Identity” (1957) as a response to this metaphysical comprehen-sion of identity. Here, Heidegger develops his notion of a belonging-together of man and be-ing, as a Sameness grounded in an enduring of the ontological difference between being and beings. In holding out this difference, man discovers that his primary belonging is that to the very being of his existence as such, rather than beings of subjective or objective kind. The no-tion of belonging-together is closely connected to the notion of the event (Ereignis), signify-ing Sameness as occurrence. The study covers Butler’s central writings up to 2006, Hegel’s Encyclopedia Logic, Science of Logic and Phenomenology of Spirit, as well as the whole of Heidegger’s late thought on this topic from the 30’s and onward together with a selection of his earlier works, among them Being and Time.
