The idiotypic network theory (INT) was conceived by the Danish immunologist Niels Kaj Jerne in 1973/1974. It proposes an overall view of the immune system as a network of lymphocytes and antibodies. The paper tries to offer a reconstruction of the genesis of the theory, now generally discarded and of mostly historical interest, first of all, by taking into account the context in which Jerne’s theoretical proposal was advanced. It is argued the theory challenged, in a sense, the supremacy of (...) the clonal selection theory (CST), this being regarded as the predominant paradigm in the immunological scenario. As CST found shortcomings in explaining certain phenomena, anomalies, one could view INT as a competing paradigm claiming to be able to make sense of such phenomena in its own conceptual framework. After a summary outline of the historical background and some relevant terminological elucidations, a narrative of the various phases of elaboration of the theory is proposed, up to its official public presentation. The concept of predisposition in medicine is ancient, and the term diathesis was used to express it since the days of Hippocrates and, especially, of Galen.The concept of diathesis was enormously popular throughout the nineteenth century, despite the vagueness of its actual meaning. It was clarified only in the early years of the twentieth century (1902), when it was however losing its clinical relevance, by a replacement of the concept ofchemical individuality by A.E. This paper is a reconstruction of Levinas’ reading of Hegel and his understanding of violence. Combining Franz Rosenzweig’s reflections which concern the sick philosopher and Hegel’s state, as well as Derrida’s interpretation of the different attributes of violence, our aim is also to give full evidence of Derrida’s critical reading of Levinas. The first part illustrates the various classifications of the figures of violence from the different periods of Hegel’s life and the traces that these figures have left in Levinas’ (...) texts beginning with ‘Liberté et commandement’ in 1953. In the second part we discuss Hegel’s well-known analogy from his Rechtsphilosophie on sovereignty and the organism—that is to say the parallel reading of some paragraphs of Naturphilosophie too—and the relation between totality and violence, in Levinas’ ‘ontology as allergy’ and in Derrida’s autoimmunology. During the 1940s and 1950s, the Australian microbiologist F. Macfarlane Burnet sought a biologically plausible explanation of antibody production. In this essay, we seek to recover the conceptual pathways that Burnet followed in his immunological theorizing. In so doing, we emphasize the influence of speculations on individuality, especially those of philosopher Alfred North Whitehead; the impact of cybernetics and information theory; and the contributions of clinical research into autoimmune disease that took place in Melbourne. We point to the influence of (...) local experimental and intellectual currents on Burnet’s work. Accordingly, this essay describes an arc distinct from most other tracings of Burnet’s conceptual development, which focus on his early bacteriophage research, his fascination with the work of Julian Huxley and other biologists in the 1920s, and his interest in North Atlantic experimental investigations in the life sciences. No doubt these too were potent influences, but they seem insufficient to explain, for example, Burnet’s sudden enthusiasm in the 1940s for immunological definitions of self and not-self. We want to demonstrate here how Burnet’s deep involvement in philosophical biology – along with attention to local clinical research – provided him with additional theoretic tools and conceptual equipment, with which to explain immune function. Ludwik Fleck is widely recognized as a precursor of Science and Technology Studies, but his case study on the development of the Wassermann reaction as a test for detecting syphilis has never been subjected to detailed empirical scrutiny. The fact that Fleck?s monograph is based on a limited set of documentary sources makes his work vulnerable to uncharitable critics. The problematic relation between thought collective and individual scientists in Fleck?s theoretical approach is another reason for a systematic re-examination of his (...) case study, using materials on the early period in the history of the Wassermann reaction (1906?1912). My re-examination highlights several problems in Fleck?s account: a misinterpretation of the switch from antigen detection to antibody detection; a neglect of the ?clinical connection?; an overemphasis on the importance of collective experience leading to implausible views on gross retrospective distortions supposedly inflicted by this experience upon the memories of individual participants; and, finally, a misjudgement of the significance of the acrimonious dispute over the intellectual ownership of the Wassermann reaction. What remains unscathed is Fleck?s picture of a zig-zag course of development from false initial assumptions via detours and cul-de-sacs to a clinically usable test in the end. (shrink)

Biological boundaries are important because of what they reveal about the evolution of a lineage, the relationship between organisms of different lineages, the structure and function of particular subsystems of the organism, the interconnection between an organism and its environment, and a myriad of other important issues related to individuality, development, and evolution. Since there is no single unifying theory for all biological sciences, there are various possible theoretical characterizations of what counts as a biological boundary. Theoretical specificity is crucial (...) for the full characterization of a boundary; to this end, it is useful to begin with general properties and then to compare these to those properties specific to a particular biological boundary. This dual heuristic approach—top-down and bottom-up—can then be repeated to increase the robustness of the boundary characterization. In what follows, I explore both the general structural and functional aspects of biological boundaries and those specific to a biological subsystem, which itself is critical to boundary formation, maintenance, and evolution—the vertebrate immune system. It is a remarkable and sobering regularity of nature that organisms remain whole because their parts are constantly being built up and broken down, and this is no less true of their boundaries. The ArgumentTwo research traditions in immunology, supposedly centered on the same issue of immune identification, have followed different theoretical goals and originated form competing phillosophical foundations. these may be labelled modernist and postmodernist, respectively, thereby applying cultural and philosophical categories to immunology in order to articulate potential scientific resonances with the broader culture. To accept that exercise and important caveat is imposed, namely, this translation is most appropriately discussed at the level of metaphor. In other words, I will structure my (...) treatment of these issues as expressed in the metaphorical language of the disipline, and thus the bulk of this discussion will focus on how the language and modeling of the science draws from the culture-at-large. Scientists seek images from their eveir everyday lives to describe phenomena that may be poorly articulated in their technical discourse; such is the utility and importance of metaphors generally, and thus it is not surprising that we might discern echoes of a postmodernist sentiment in the metaphors borrowed from post–World War II culture. I Will also discuss, to a more limited extent, how postmodernists have sought support for their own ideological arguments in immunology. This last topic server only to illustrate the bidirectionality of scientific discourse with the society in which it is embedded. (shrink)

Few topics in contemporary science hold the wide interest commanded by immunology, so this graceful and timely account of the development of this science is a welcomed addition to the literature. Succinct, well-written, and informed, Intolerant Bodies narrates the history of immunology through the lens of autoimmune disease. In what the authors call “a biography” , they have focused on four central illnesses: multiple sclerosis, systemic lupus erythematosus, rheumatoid arthritis, and type 1 diabetes mellitus. However, the story told here extends (...) far beyond the topic of “attack against self” to provide perhaps the best overview of immunity available for the general reader. The specialist will also appreciate the rich anecdotal material and benefit from the historical insights scattered throughout the text. So the book is, in fact, more than a history of autoimmunity and offers a compact history for those who need not consult more comprehensive accounts (e .. (shrink)

It has long been taken for granted that the immune system’s capacity to protect an individual from infection and disease depends on the power of the system to distinguish between self and nonself. However, accumulating data have undermined this fundamental concept. Evidence against the self/nonself discrimination model left researchers in need of a new overarching framework able to capture the immune system’s reactivity. Here, I highlight that along with the self/nonself model, another powerful representation of the immune system’s reactivity has (...) been developed in the twentieth century immunology. According to this alternative view, the immune system is not a killer of nonself strangers but a peace-maker helping to establish harmony with the environment. The balance view of the system has never become part of the dominant paradigm. However, it is gaining more and more currency as new research develops. Advances in mucosal immunology confirm that instead of distinguishing between self and foreign the immune system reacts to microbial, chemical and self-induced alterations to produce responses that counterbalance effects of these changes. Originating in the work of Ernst Haeckel and Wilhelm Preyer, and advanced by a Prussian embryologist, Wilhelm Roux, the idea of struggle for existence between body parts helped to establish a framework, in which population cell dynamics rather than a predefined harmony guides adaptive changes in an organism. Intended to provide a causal-mechanical view of functional adjustments in body parts, this framework was also embraced later by early pioneers of immunology to address the question of vaccine effectiveness and pathogen resistance. As an extension of these early efforts, Elie Metchnikoff established an evolutionary vision of immunity, development, pathology, and senescence, in which phagocyte-driven selection and struggle promote adaptive changes in an organism. Despite its promising start, the idea of somatic evolution lost its appeal at the turn of the twentieth century giving way to a vision, in which an organism operates as a genetically uniform, harmonious entity. How does one testify and, moreover, testify philosophically to the experience of receiving an organ transplant? What kinds of survival or forms of living are being fostered by newly emerging conjunctions between philosophy and biomedicine? Focusing on transplantation and immunology, we are going to reflect on some of the ways and styles in which motifs drawn from these biomedical fields have come to occupy an increasingly prominent place in recent philosophy expressing and formulating different concerns and paradigms. Varela and Jean-Luc Nancy's autobiographical reflections on transplantation, we are going to consider distinct moments and preoccupations at work in the motifs of immunity, autoimmunity, the pharmakon and the graft in the thought of Jacques Derrida, Roberto Esposito and Bernard Stiegler. ArgumentDiphtheria serum production in France was dominated by the Pasteur Institute, which equipped a facility at Garches to produce the antitoxin on a large scale. This article treats the background to the founding of this facility, as well as its day-to-day functioning around 1900. The treatment integrates an examination of the practical undertaking of serum production by the Pasteur Institute with an analysis of the popular perception of the Institute and the mixed financing of the whole venture. We particularly emphasize (...) the “industrial” features of this manufacturing process that involved living units of production, showing how bioassays influenced the destiny of the animals producing the serum. Finally, we argue that this monitoring of the horses, seen as serum-producing units, also provided information on the diseases the sera were intended to treat. (shrink)

Today, scientific biography is primarily thought of as a way of writing contextual history of science. But the genre has other functions as well. This article discusses seven kinds of ideal–typical subgenres of scientific biography. In addition to its mainstream function as an ancilla historiae, it is also frequently used to enrich the understanding of the individual construction of scientific knowledge, to promote the public engagement with science, and as a substitute for belles-lettres. Currently less acknowledged kinds of scientific biography (...) include its use as a medium for public and private, respectively, commemoration. Finally, the use of scientific biography as a research ethical genre, providing examples of ‘the good life in science’, is emphasized. (shrink)

In this paper, I revisit a problem in immunology and molecular genetics that I had first tried to understand some philosophical implications of over twenty years ago. At that time, some immunologists such as Mel Cohn at the Salk Institute, referred to it as the GOD problem, which was the acronym forGeneratorOfDiversity (also see Cohn's more recent discussion in his 1994, 41-48). In the early 1970s there were three or four different theories that had been proposed to account for the (...) way in which antibody diversity is generated and considerable argument among the proponents of the different approaches to the problem (see Cohn, 1994). In 1971-72 I attempted to assess the strengths and weaknesses of the competing theories of antibody diversity from the perspective of a “logic of comparative theory evaluation” that had seemed to work reasonably well in physics (Schaffner, 1970), and I drafted an essay that ultimately sided with one of those theories (somatic mutation). In this article I consider some central aspects of the naturalist philosophy of science and science and technology studies in dealing with the contested status of technoscience in medicine. Focusing on the concepts of realism and representation, I argue that theories of science-as-practice in naturalist philosophy of science should expand their scope so as to reflect more thoroughly on the social and political context of technoscience. I develop a hermeneutic of technomedical objects in order to highlight the internal connectedness between (...) social action, material agency, and the actions of scientific communities. The framework thus developed is used to re-consider the genomic turn in medicine. Pointing to the discrepancies between socially dominant representations of genomic technology and the actual interventions brought about through those technologies, I raise the question of how and where to address problems of theory and policy. The present article examines the material, epistemological, and social dimensions of late nineteenth-century anti-cholera serum controversies that unfolded in Tokyo and Berlin. It seeks to shed light on the conflicting values embedded in the construction of scientific evidence during the transnational exploration of bacteriology as an effective response to controlling epidemics. Driven by Japanese health authorities’ initiatives, Japanese bacteriologist Kitasato Shibasaburo participated in the elaboration of bacteriological research oriented toward therapeutic application during his stay in Berlin. This work coincided with (...) the rise of a controversy over anti-cholera serum in relation to the animal experiments conducted by Richard Pfeiffer, a German bacteriologist. After presenting a series of animal experiments and certain clinical cases conducted in Germany, France, and Egypt in the context of the controversies, the article analyzes a therapeutic trial conducted by Kitasato in Japan during an 1895 cholera epidemic. His strategy, bringing new data to the transnational debate through an intensive investment of resources in the trial, provoked criticism from his Japanese and German colleagues. These critics questioned Kitasato’s method and pointed out the low social value of this experimental serum therapy, performed in highly equipped conditions. Through this case study, the present article highlights: a) a strong tension between transnational research interactions among bacteriologists and local medical practices during public health campaigns against epidemics at the turn of the twentieth century; b) the importance of analyzing the interconnected effects of local, national and transnational frameworks of medical science when examining the increasingly intertwined relationships between laboratory science and clinical medicine. Biomedical researchers claim there is significant biomedical information about humans which can be discovered only through experiments on intact animal systems (AMA p. 2). Although epidemiological studies, computer simulations, clinical investigation, and cell and tissue cultures have become important weapons in the biomedical scientists' arsenal, these are primarily "adjuncts to the use of animals in research" (Sigma Xi p. 76). Controlled laboratory experiments are the core of the scientific enterprise. Biomedical researchers claim these should be conducted on intact biological systems, (...) whole animals. By observing the effects of various stimuli in non-human animals, we can form legitimate expectations about the likely effects of these stimuli in humans. A longstanding philosophical premise perceives simplicity as a desirable attribute of scientific theories. One of several raised justifications for this notion is that simple theories are more likely to indicate the true makeup of natural systems. Qualitatively parsimonious hypotheses and theories keep to a minimum the number of different postulated entities within a system. Formulation of such ontologically simple working hypotheses proved to be useful in the experimental probing of narrowly defined bio systems. It is less certain, however, whether qualitatively (...) parsimonious theories are effective indicators of the true nature of complex biological systems. This paper assesses the success of ontologically simple theories in envisaging the makeup of three complex systems in bacteriology, immunology, and molecular biology. Evidence shows that parsimonious theories completely misconstrued the actual ontologically complex constitutions of the three examined systems. Since evolution and selective pressures typically produce ontologically intricate rather than simple bio systems, qualitatively parsimonious theories are mostly inapt indicators of the true nature of complex biological systems. ArgumentThe experimental development of a therapeutic serum against diphtheria between 1891 and 1894 was characterized by a scientific competition that pitted Emil Behring from the Institute for Infectious Diseases in Berlin against Émile Roux and Elie Metschnikoff from the Pasteur Institute in Paris. In general, their competition can be regarded as an extension of the fundamental differences that separated the research schools of Robert Koch and Louis Pasteur. However, to characterize the competition for a diphtheria-serum as “national rivalry” fails to (...) account adequately for the mutual adoption of experimental practices by the Berlin and Parisian protagonists, whose contributions to the development of a therapeutic serum were intertwined in complex ways. Nor can it be characterized as “cooperation,” given their fierce public disputes over scientific concepts and the fact that these disputes also shaped the peculiarities of the experimental procedures in Berlin and Paris. A close analysis reveals a complex picture of the dynamic interaction between the conceptual and experimental activities of Behring, Roux, and Metschnikoff – interaction that defined as well as bridged the “French” and “Prussian” experimental systems of diphtheria-serum research. ArgumentThe procedure ofWertbestimmungplayed a vital role in the implementation of serum therapy and the standardization of mass-produced pharmaceuticals. In fin-de-siècle Germany, a legal framework was put in place to guarantee serum quality and safety and to minimize any associated public health risks. Because the sera were biological remedies, it was difficult to produce them in uniform quality and the procedure ofWertbestimmung, i.e. determining the potency of the serum based on an objective and comparable value, was extremely complex. Various agents such (...) as bacteria cultures, serum hosts, or test animals had to be regulated. In the years after 1895, numerous efforts to stabilize the procedures ofWertbestimmungwere undertaken by serum producers and members of the state-run survey institute responsible for overseeing serum production. Despite efforts to stabilize the framework and to generate a reliable reference system, the framework's environment and agents were in constant flux: new producers entered the market and procedures were expanded to include other biologicals as well. The article describes the dynamics involved in the sustained efforts to maintain a stable framework in the face of constant alterations between 1895 and the 1920s. (shrink)

ArgumentThis essay places some therapeutic vaccines, including particularly the diphtheria antitoxin, into their larger historical context of the late nineteenth century. As industrially produced drugs, these vaccines ought to be seen in connection with the structural changes in medicine and pharmacology at the time. Given the spread of industrial culture and technology into the field of medicine and pharmacology, therapeutic vaccines can be understood as boundary objects that required and facilitated communication between industrialists, medical researchers, public health officials, and clinicians. (...) It was in particular in relation to evaluation and testing for efficacy in animal models that these medicines became a model for twentieth-century medicine. In addition, these medicines came into being as a parallel invention in two very distinct local cultures of research: the Institut Pasteur in Paris and the Institut für Infektionskrankheiten in Berlin. While their local cultural origins were plainly visible, the medicines played an important role in the alignment of the methods and objects that took place in bacteriology research in France and Germany in the 1890s. This article assesses the two locally specific regimes for control in France and in Imperial Germany. In France the Institut Pasteur, building on earlier successful vaccines, enjoyed freedom from scrutinizing control. The tight and elaborate system of control that evolved in Imperial Germany is portrayed as being reliant on experiences that were drawn from the dramatic events that surrounded the launching of a first example of so-called “bacteriological medicine,” tuberculin, in 1890. This essay examines the role of social interactions in the search for blood stem cells, in a recent episode of biomedical research. Linked to mid-20th century cell biology, genetics and radiation research, the search for blood stem cells coalesced in the 1960s and took a developmental turn in the late 1980s, with significant ramifications for immunology, stem cell and cancer biology. Like much contemporary biomedical research, this line of inquiry exhibits a complex social structure and includes several prominent scientific successes, (...) recognized as such by participating researchers. I use personal interviews and the published record to trace the social interactions crucial for scientific success in this episode. All recognized successes in this episode have two aspects: improved models of blood cell development, and new interfaces with other lines of research. The narrative of the search for blood stem cells thus yields a robust account of scientific success in practice, which generalizes to other scientific episodes and lends itself to expansion to include wider social contexts. The article discusses Karl Popper’s account of Lamarckism. In this article I use Popper’s published and unpublished statements regarding Lamarckism as well as his correspondence with the Australian immunologist Edward Steele and other biologists to examine why Popper was interested in Lamarckism, how his account of Lamarckism can be understood in the context of his philosophy, and what, if any, new context Popper provided for the discussion of this abandoned doctrine. I begin by discussing Popper’s frame of reference regarding Lamarckism, (...) examining how it changed following his involvement with different “transitory groups” of biologists concerned with redefining the boundaries between traditional disciplines. I then discuss Popper’s account of Lamarckism in positioning it in relation to three aspects of his philosophy: his distinction between the principles of induction/instruction and deduction/selection, his distinction between scientific and teleological concepts, and his concept of progress in science. Then, I follow the debate concerning Steele’s Lamarckian experiments with a closer look at Popper’s role and his position in this controversy. I conclude that Popper’s writings provided a new context for the discussion of Lamarckism in the 1970s and 1980s, thus making it “scientifically acceptable” again. I preface this discussion by a brief outline of Steele’s Lamarckian hypothesis and Popper’s comments on Steele’s work. (shrink)

In 1957, Francis Crick outlined a startling vision of life in which the great diversity of forms and shapes of macromolecules was encoded in the one-dimensional sequence of nucleic acids. This paper situates Crick's new vision in the debates of the 1950s about protein synthesis and gene action. After exploring the reception of Crick's ideas, it shows how they differed radically from a different model of protein synthesis which enjoyed wide currency in that decade. In this alternative model, advocated by (...) Linus Pauling and other luminaries, three-dimensional templates directed the folding of proteins. Even though it was always considered somewhat speculative, this theory was supported by a number of empirical results originating in different experimental systems. It was eventually replaced by a model in which the forms and shapes of macromolecules resulted solely from their amino acid sequence, dramatically simplifying the problem of protein synthesis which Crick was attempting to solve in 1957. The historiography of immunology since 1999 is reviewed, in part as a response to claims by historians such as Thomas Söderqvist the field was still immature at the time. First addressed are the difficulties, past and present, surrounding the disciplinary definition of immunology, which is followed by a commentary on the recent scholarship devoted to the concept of the immune self. The new literature on broad immunological topics is examined and assessed, and specific charges leveled against the paucity of certain (...) types of histories, e.g. biographical and institutional histories, are evaluated. In conclusion, there are compelling indications that the history of immunology has moved past the initial tentative stages identified in the earlier reviews to become a bustling, pluripotent discipline, much like the subject of its scrutiny, and that it continues to develop in many new and exciting directions. The objective of this introduction is to outline certain historical and theoretical frames of modern immunology. Immunology as a discipline that has been shaped relatively recently attempts to explain all kinds of phenomena safeguarding the integrity of an organism. An organism is understood as a complex multi-system structure developed in the course of evolution. In the case of such a broad sense concept of immunity a penetrating discussion is required. Thus, biology philosophers as well as scientists – immunologists, for nearly (...) 50. years have been analysing the basic definitions, which led them to interesting conclusions. (shrink)

Najpierw wskazano przeszłe i teraźniejsze problemy, które historiografia ma ze zdefiniowaniem immunologii, a następnie skomentowano ostatnie studia nad pojęciem immunologicznego „ja”. W dalszym toku przeglądu przeanalizowano i oceniono nowe publikacje poświęcone zróżnicowanym zagadnieniom immunologii oraz niektóre charakterystyczne oskarżenia formułowane wobec niedostatku pewnych dziedzin historii, (...) na przykład historii biograficznych i historii instytucji. W podsumowaniu przedstawiono konkurujące ze sobą kierunki, w których rozwinęła się historia immunologii, wychodząc poza wstępne, tymczasowe stadia uchwycone w poprzednim przeglądzie, i stając się wielogłosową, pluripotencjalną dyscypliną, podobną do przedmiotu jej badań. The dissertation criticizes two analogical applications of Darwinism to the spheres of mind and culture: the Darwinian approach to creativity and memetics. These theories rely on three basic analogies: the ontological analogy states that the basic ontological units of culture are so-called memes, which are replicators like genes; the origination analogy states that novelty in human creativity emerges in a "blind" Darwinian manner; and the explanatory units of selection analogy states that memes are "egoistic" and that they can spread independently (...) of our mind. The detailed philosophical analysis offered in the dissertation shows that these three analogies rely on either wrong or trivial statements: they provide either wrong or no new descriptions or explanations of the phenomena at hand. In Chapter One, I introduce the diverse ways, in which contemporary Darwinism is used today outside of evolutionary biology. Chapter Two explains what it means to claim that something evolves in a Darwinian manner. Chapter Three to Five address each of the three analogies separately. Darwinism applied to culture and mind, at least in the way discussed in this study, is not a dangerous idea. It leads either to wrong claims or to a re-telling in Darwinian terms of what we have already known. (shrink)
