Today, the lipid bilayer structure is nearly ubiquitous, taken for granted in even the most rudimentary introductions to cell biology. Yet the image of the lipid bilayer, built out of lipids with heads and tails, went from having obscure origins deep in colloid chemical theory in 1924 to being “obvious to any competent physical chemist” by 1935. This chapter examines how this schematic, strictly heuristic explanation of the idea of molecular orientation was developed within colloid physical (...) chemistry, and how the image was transformed into a reflection of the reality and agency of lipid molecules in the biological microworld. Whereas in physical and colloid chemistry these images considered secondary to instrumental measurement and mathematical modeling of surface phenomena, in biology the manipulable image of the lipid on paper became an essential tool for the molecularization of the cell. Abstract: The analytical, comparative cross-sectional study was conducted to assess the thyroid profiles and modified lipid profiles levels among Sudanese patients with coronary heart disease performed on forty-one patients with coronary heart disease as test group collected from Sudan Heart Center, Al rebat teaching hospital and Al mawada hospital in Khartoum state, during the period between November 2017 and May 2018. Furthermore, the test group compared with forty-one apparently healthy volunteers as control group was selected with the same inclusion (...) criteria. Spectrophotometeric methods were used for measurement of lipid profile. Thyroid hormones (T3 &T4) and thyroid stimulating hormone were measured by using Sandwich Enzyme-Linked Immunosorbent Assay (ELISA). Moreover, statistical package for social science (SPSS version 23) computer software was used for data analysis. The study clearly demonstrated that there was statistically significant decrease in T3, and significant increase in T4 levels in test group compared to control with (P. value 0.047) for T3, and (P. value 0.002) for T4.Nonetheless, the result of this study indicated a significant elevation in mean of total cholesterol, low density lipoprotein-cholesterol levels and significant decrease in high density lipoprotein –cholesterol in test group compared to control (P.value 0.000). In conclusion Coronary heart diseases patients have significantly low Triiodothyronine, elevated Thyroxin, and normal Thyroid Stimulating Hormone levels, with significantly higher Total Cholesterol, Low Density Lipoprotein-Cholesterol concentrations, and lower HighDensity Lipoprotein-Cholesterol concentrations. • Aggregates of previously isolated cells of Hydra are capable, under suitable solvant conditions, of regeneration forming complete animals. In a first stage, ecto- and endodermal cells sort out, producing the bilayered hollow structure characteristic of Hydra tissue; thereafter, heads are formed (even if the original cell preparation contained no head cells), eventually leading to the separation of normal animals with head, body column and foot. Hydra appears to be the highest type of organism that allows for regeneration of the (...) entire structure from random cell aggregates. The system is particularly useful for studying cell interactions, tissue polarity, pattern formation, and cell differentiation. (shrink)

The environment can elicit biological responses such as oxidative stress (OS) and inflammation as a consequence of chemical, physical, or psychological changes. As population studies are essential for establishing these environment-organism interactions, biomarkers of OS or inflammation are critical in formulating mechanistic hypotheses. By using examples of stress induced by various mechanisms, we focus on the biomarkers that have been used to assess OS and inflammation in these conditions. We discuss the difference between biomarkers that are the result of a (...) chemical reaction (such as lipid peroxides or oxidized proteins that are a result of the reaction of molecules with reactive oxygen species) and those that represent the biological response to stress, such as the transcription factor NRF2 or inflammation and inflammatory cytokines. The high-throughput and holistic approaches to biomarker discovery used extensively in large-scale molecular epidemiological exposome are also discussed in the context of human exposure to environmental stressors. We propose to consider the role of biomarkers as signs and to distinguish between signs that are just indicators of biological processes and proxies that one can interact with and modify the disease process. The temptation for male hunters, far from any home base, to eat the best portions of meat at the kill (...) site — as do other social carnivores — called for strong measures from human females, who were paying the heavy metabolic and physical costs of bearing large-brained but helpless children. Even in the modern west, with well stocked supermarkets, a pregnant or lactating woman can lose ten percent of the dry weight of her brain, because developing babies demand dietary lipids for brain growth (Horrobin, 1998). Hence the idea of the menstrual sex strike, designed to force males to deliver their kills entirely into the hands of women for cooking and distribution—a practice common in foraging communities to this day. Objective: Laparoscopic ovarian drilling normalizes ovulation by reducing cortical thickening, lowering androgen production, and regulating luteinizing hormone pulse frequency. On the other hand, the effect of laparoscopic ovarian drilling on the endometrium is unknown. This study was planned to investigate the changes in the functional markers of choline, creatine, lactate, and lipid metabolites of the endometrium before and after laparoscopic ovarian drilling in women with polycystic ovary syndrome. -/- Methods: Twenty women diagnosed with clomiphene-resistant polycystic ovary syndrome who did (...) not ovulate successfully despite the administration of clomiphene citrate or aromatase inhibitor were included in the study. Patients were offered Assisted Reproductive Technology (ART) or laparoscopic ovarian drilling options. Patients who accepted laparoscopic ovarian drilling formed the study group. Endometrial MR spectroscopy was applied to the participants in the mid-luteal phase before laparoscopic ovarian drilling. Choline, creatine, lactate, and lipid metabolites of all patients were measured and denominated parts per million. The second MR spectroscopy was performed 2 months after the first MR spectroscopy, and the changes in endometrial metabolites after laparoscopic ovarian drilling were recorded. Twenty patients who did not have clinical and laboratory findings of polycystic ovary syndrome and were matched for age and body mass index were accepted as the control group. The patients in this group were selected from fertile women with at least 2 children. MR spectroscopy was performed in the mid-luteal phase in fertile women. The obtained results were compared within and between groups and the possible effects of laparoscopic ovarian drilling on metabolite synthesis were tried to be determined. -/- Results: During MR spectroscopy examination main endometrial metabolites choline, creatine, lactate, and lipid were detected in the polycystic ovary syndrome group. The most prominent metabolite peak before and after laparoscopic ovarian drilling was recorded as choline and creatine. There was a significant increase in choline and creatine peaks after laparoscopic ovarian drilling compared to the values before laparoscopic ovarian drilling. There was no significant increase in lactate and lipid signals before and after laparoscopic ovarian drilling. The choline and creatine metabolite levels of the women with polycystic ovary syndrome before laparoscopic ovarian drilling were significantly lower than those of the fertile women. The choline and creatine metabolite levels of the women with polycystic ovary syndrome after m laparoscopic ovarian drilling were similar to those of the fertile women. There was no significant difference between lactate and lipid signals before and after laparoscopic ovarian drilling. -/- Conclusion: Laparoscopic ovarian drilling improves polycystic ovary syndrome-related subfertility by increasing endometrial choline and creatine metabolite levels to those of fertile women. Introduction: Metabolic syndrome (MetS) is an endocrinopathy with a combination of cardiovascular and metabolic compounds. In our study, it is expected to obtain results showing that mortality rate, loss of workforce, and treatment costs due to disorders caused by MetS can be reduced by physical exercise. The study analyses the effect of moderate exercise training on this Apolipoprotein E (ApoE), Apolipoprotein CIII (ApoCIII), adiponectin, resistin, interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α) which are thought to have a role in the (...) deterioration of glucose and lipid metabolism in MetS. -/- Methods: This clinical experimental study consists of 3 groups. The MetS+E (n=24) group, which included the participants who agreed to participate in the exercise program in addition to their medical treatment, the MetS (n=23) group who received medical treatment but did not exercise, and the Control+E (n=25) group, which included healthy volunteers who had the same protocol as MetS+E ApoE, ApoCIII, adiponectin, resistin, IL-6, and TNF-α plasma levels of all participants were measured both at the beginning of the study and at the end of the protocol. -/- Results: At the end of the study we reached the following findings; insulin and Homeostatic Model Assessment for Insulin Resistance (HOMA-IR) levels decreased in exercise groups (p=0,03). ApoCIII levels increased in all the groups after the study (p<0,01). IL-6 levels decreased in MetS+E (p<0,01) and Control+E (p=0,037). ApoE (p=0,01) and TNF-α (p=0,037) levels decreased only in the Control+E group. -/- Conclusion: Training showed metabolic, anti-inflammatory, and physical improvements independent of ApoE and ApoCIII in those with MetS. Different anesthetics are known to modulate different types of membrane-bound receptors. Their common mechanism of action is expected to alter the mechanism for consciousness. Consciousness is hypothesized as the integral of all the units of internal sensations induced by reactivation of inter-postsynaptic membrane functional LINKs during mechanisms that lead to oscillating potentials. The thermodynamics of the spontaneous lateral curvature of lipid membranes induced by lipophilic anesthetics can lead to the formation of non-specific inter-postsynaptic membrane functional LINKs by different mechanisms. (...) These include direct membrane contact by excluding the inter-membrane hydrophilic region and readily reversible partial membrane hemifusion. The constant reorganization of the lipid membranes at the lateral edges of the postsynaptic terminals (dendritic spines) resulting from AMPA receptor-subunit vesicle exocytosis and endocytosis can favor the effect of anesthetic molecules on lipid membranes at this location. Induction of a large number of non-specific LINKs can alter the conformation of the integral of the units of internal sensations that maintain consciousness. Anesthetic requirement is reduced in the presence of dopamine that causes enlargement of dendritic spines. Externally applied pressure can transduce from the middle ear through the perilymph, cerebrospinal fluid, and the recently discovered glymphatic pathway to the extracellular matrix space, and finally to the paravenular space. The pressure gradient reduce solubility and displace anesthetic molecules from the membranes into the paravenular space, explaining the pressure reversal of anesthesia. Changes in membrane composition and the conversion of membrane hemifusion to fusion due to defects in the checkpoint mechanisms can lead to cytoplasmic content mixing between neurons and cause neurodegenerative changes. The common mechanism of anesthetics presented here can operate along with the known specific actions of different anesthetics. (shrink)

Perception is a first-person internal sensation induced within the nervous system at the time of arrival of sensory stimuli from objects in the environment. Lack of access to the first-person properties has limited viewing perception as an emergent property and it is currently being studied using third-person observed findings from various levels. One feasible approach to understand its mechanism is to build a hypothesis for the specific conditions and required circuit features of the nodal points where the mechanistic operation of (...) perception take place for one type of sensation in one species and to verify it for the presence of comparable circuit properties for perceiving a different sensation in a different species. The present work explains visual perception in mammalian nervous system from a first-person frame of reference and provides explanations for the homogeneity of perception of visual stimuli above flicker fusion frequency, the perception of objects at locations different from their actual position, the smooth pursuit and saccadic eye movements, the perception of object borders, and perception of pressure phosphenes. Using results from temporal resolution studies and the known details of visual cortical circuitry, explanations are provided for (a) the perception of rapidly changing visual stimuli, (b) how the perception of objects occurs in the correct orientation even though, according to the third-person view, activity from the visual stimulus reaches the cortices in an inverted manner and (c) the functional significance of well-conserved columnar organization of the visual cortex. A comparable circuitry detected in a different nervous system in a remote species-the olfactory circuitry of the fruit fly Drosophila melanogaster-provides an opportunity to explore circuit functions using genetic manipulations, which, along with high-resolution microscopic techniques and lipid membrane interaction studies, will be able to verify the structure-function details of the presented mechanism of perception. The present investigation has been designed to study the effect of sub lethal concentrations of Nickel on the serum metabolites of Channa punctatus after exposure to 45 days. The present study shows that serum metabolites such as glucose and protein were significantly decreased while lipid was significantly increased. Thus, the result provides an overview of the manipulation of fish, Channa punctatus as a biomarker of heavy metals through alternation in serum biochemical parameters.

ALTERAÇÕES DO CORPO LÚTEO -/- A estrutura funcional desenvolve-se a partir da cavidade folicular após a ovulação. O corpo lúteo é constituído pelas células da teca interna (pequenas e ativas na primeira fase do seu desenvolvimento) e células da granulosa (grandes e ativas na segunda metade do seu (...) desenvolvimento). As principais características de cada tipo de célula podem resumir-se da seguinte forma: As células pequenas são mais sensíveis à ação da LH porque possui uma quantidade maior de receptores biologicamente ativos para esta hormona e a resposta é mediada pelo sistema AMPc-adelinato ciclase. As grandes células lúteas têm menos receptores para a LH (a maior quantidade são para a PGF2α e a PGE2), e são as responsáveis pela produção da oxitocina luteal (BAH, et al. 2006; CAMPO, 2003). Contrariamente às pequenas células lúteas, a produção de progesterona (P4) é parcialmente controlada pelo sistema Ca+2 - Fosfatil inositol-quinase C (CAMPO, 2003). A regulação da função do corpo lúteo é um mecanismo complexo. Nela intervêm fatores tróficos e líticos que estão presentes em forma simultânea durante todo o ciclo estral, e portanto a estimulação ou inibição da síntese e secreção de P4 depende do balanço entre estes fatores. (CAMPO, 2003). Dentro dos fatores tróficos, os nutrientes desempenham um papel relevante sobre a luteólise e as funções luteotrópicas, entre estas pode-se citar a ação das seguintes: -/- 7.1 Vitaminas Deficiência em betacarotenos. Vitamina A é um termo genérico que designa qualquer composto com atividade de retinol, o trans-retinol, é o composto base, sua forma aldeído e ácido são chamados de retinol e ácido retinóico, respectivamente. Por outro lado, a forma ativa da Vitamina A participa no mecanismo da visão, como 11-cis-retinal. Adicionalmente, o betacaroteno, considerado uma provitamina A é a única forma em que esta encontra nas plantas. Em sentido nutricional, a família da Vitamina A compreende todos os compostos que possuem atividade de retinol, incluindo cerca de cinquenta carotenoides que possuem atividade de provitamina A, aceitando-se que o mais ativo é o trans-betacaroteno (TORRES, et al. 2002). As vitaminas que são necessárias para o crescimento e a produção, também são essenciais para a reprodução. As recomendações atuais de vitamina A são de 75.000 UI/dia durante a lactação e 80.000 UI/dia (nota: 1 mg de Vitamina A = 400 UI) durante o período seco nas vacas leiteiras. A suplementação com vitamina A tem pouco risco de toxicidade, enquanto a sua deficiência pode retardar o aparecimento do primeiro cio/estro, induzir cios/estros silenciosos, aumentar o número de quistos ováricos, reduzir os índices de concepção, causar abortos, mortes embrionárias e ocasionar o nascimento de bezerros debilitados e fracos, uma vez que não irão nascer com os índices e requerimentos ideais de nutrientes passados da mãe para o feto pela placenta, e que, já que a mãe possui deficiência nutricional, o colostro não terá o efeito completo para passar a imunidade essencial ao neonato. Embora os mecanismos de ação não sejam claros, foram notificadas maiores secreções de esteroides ováricos quando as concentrações de β-carotenos no fluido folicular foram elevadas; logo, foram observadas maiores concentrações elevadas de β- estradiol e vitamina A nos folículos que terminaram em ovulação do que naqueles que terminaram em atresia. Para evitar as perdas reprodutivas por deficiência de vitamina A, pode-se optar pela administração oral ou injetável desta ao animal, mediante dois métodos: o primeiro consiste em adicionar 1 a 2,5 kg de premix vitamínico em 1 tonelada de grãos o que supre de 4 a 6.000 UI o que é desejável; o segundo é a administração via injetável em quantidades vide bula do fármaco. 7.2 Minerais O excesso de fósforo é absorvido principalmente no intestino delgado dos mamíferos através de um mecanismo de transporte ativo dependente do sódio, que é estimulado pela pró-hormona 1,25- hidroxivitamina D3. Os ruminantes são capazes de absorver o fósforo dietético em proporção direta às quantidades presentes na ração. Estima-se que aproximadamente 1% do corpo animal é composto de fósforo, do qual 85% está presente nos dentes e no tecido ósseo. Os tecidos moles são capazes de conter entre 0,15-0,20% de fósforo sob a forma de fosfoproteínas, nucleoproteínas, fosfolipídios, fosfato creatinina, adenosintrifosfato, glicose-6-fosfato, etc. Ao nível do plasma sanguíneo, o fósforo apresenta valores de 12,09 mg/dl. Os valores plasmáticos de fósforo são regulados principalmente pelo rim, sendo observado um aumento na excreção urinária do elemento inorgânico quando este excede a capacidade de transporte (REBOLLAR & MATEOS, 1999). O fósforo faz parte dos fosfolipídios, que são importantes no transporte de lipídios e seu metabolismo, como componente das membranas celulares. O fosfato faz parte do RNA e do DNA, componentes celulares vitais, e são essenciais para a síntese proteica. O fosfato faz parte de sistemas enzimáticos como a cocarboxilase e NAD, portanto está envolvido em todos os processos de síntese e regulação energética, dado que o corpo lúteo requer altas quantidades de energia para a síntese celular e hormonal, um excesso de fósforo pode bloquear a capacidade renal da sua regulação gerando um desequilíbrio catiônico que descompensa a relação Na:K, já que deverão ser excretados íons fosfato para regular o equilíbrio ácido-básico, desta forma, os processos celulares em tecidos de alta sensibilidade como o ovário serão afetados em sua função. A situação para prevenção de problemas relacionados ao fósforo (P) já foi tratada em assuntos anteriores, porém, sempre vale frisar para o leitor fiel que me lê que as exigências de P presentes na matéria seca (MS) variam entre 0,17 e 0,59% de P por kg/MS para bovinos de corte e em 0,32 a 0,44% por kg/MS para o gado leiteiro. Dessa forma, dados obtidos por pesquisas, ditam que para a mantença de uma vaca ou quaisquer outra categoria bovina o ideal é o consumo de 1.43 g/100 kg de peso vivo (PV), garantindo assim a prevenção de possíveis afecções reprodutivas como as alterações do corpo lúteo. Gramíneas, forragens, leguminosas e os concentrados fornecem muito bem os requerimentos de fósforo para os animais, todavia deve-se estar atento as relações e dependências do P com outros elementos como o Ca e a relação Na:K (10-15:1 idealmente). 7.3 Compostos estrogênicos Os fitoestrogênios afetam repetidamente a estabilidade do corpo lúteo. Os compostos estrogênicos encontrados principalmente em leguminosas pertencem principalmente a três grupos: isoflavanos, isoflavonas, presentes no gênero Trifolium e praticamente restritos às leguminosas, e cumestanos, que foram estudados na alfafa (Medicago sativa) e na Melilotus alba. Todas as isoflavonas sofrem algum grau de transformação no rúmen, sendo o Isoflavonoide formononetina, na realidade um pró-estrógeno, o único que se ativa no ecossistema ruminal ao ser convertido em equol, enquanto que as demais são degradadas a produtos inativos (RAMOS, et al. 1998). Por fim, a alta concentração no sangue do equol retarda a retroalimentação sobre a hipófise mediante a qual se reduzem os níveis de FSH para dar lugar à onda ovulatória de LH, se esta não ocorrer, o corpo lúteo obviamente não se formará. Un ghid complet pentru cultivarea şi prepararea celor mai variate tipuri de cafea, cu accent pe aspectele culturale şi de sănătate, şi modalităţi de includere a cafelei în diverse deserturi şi cocktailuri. Cafeaua este o băutură universal recunoscută ca o necesitate umană. Departe de a fi văzută ca un lux sau privită cu indulgenţă, ea este considerată un corolar pentru energia şi eficienţa umană, producând în acelaşi timp o puternică senzaţie de plăcere. Cafeaua este o băutură democratică. Este în acelaşi (...) timp băutura înaltei societăţi, dar şi a bărbaţilor şi femeilor care muncesc în diverse domenii de activitate, mental, sau fizic. Tocmai de aceea cafeaua a mai fost numită şi "lubrifiantul om-maşină cel mai cunoscut", ca şi "gustul cel mai plăcut din toată natura." Dar cafeaua este ceva mai mult decât o băutură. Este unul dintre cei mai importanţi adjuvanţi alimentari din lume. Există şi alte alimente auxiliare, dar niciunul care să exceleze în gust şi efecte reconfortante precum cafeaua, efecte datorate în primul rând aromei sale unice.
