Scholarship on Plato's Timaeus has paid relatively little attention to Tim. 77a–81, a seemingly disjointed passage on topics including plants, respiration, blood circulation, and musical sounds. Despite this comparative neglect, commentators both ancient and modern have levelled a number of serious charges against Timaeus' remarks in the passage, questioning the coherence and explanatory power of what they take to be a theory of respiration. In this paper, I argue that the project of 77a–81e is not to sketch theories of respiration, (...) circulation, and digestion, but to explain how the human body is maintained in light of and despite constant environmental depletion. Further, I argue that in order to understand this account of “the replenishing system,” we need to understand Timaeus' striking analogy of the fish trap or nassa. Commentators have generally focused directly on the workings of the bodily construction that Timaeus likens to a fish trap, but without considering how we should understand the analogy qua analogy. I develop a functional reading of the analogy that yields a coherent account of the replenishing system on which previous criticisms of Timaeus' remarks on respiration do not arise. Aside from lending greater unity to the passage, both internally and within its immediate context in the dialogue, this account of the replenishing system contributes to our understanding of Timaeus' reason-and-necessity explanatory framework as applied to the human body and has noteworthy implications for specific explanatory principles, in particular like-to-like motion and circular thrust. Kamm’s sore throat case gives us a choice: save one life, or save a distinct life and cure a sore throat. We defend the fairness explanation of the judgement that one should flip a coin to decide whom to save: it is disrespectful to let a sore throat act as a tie-breaker, because an individual would be forced to forgo a 50% fair chance of living (given to them by a coin flip), which cannot be outweighed by any number of (...) sore throats. We show that this explanation of when and why claims can permissibly break ties generates new problem cases for theories of aggregation, including theories that have been thought to accommodate the judgement that one should flip a coin in Kamm’s sore throat case. We then generalise the fairness explanation to cases involving multiple groups. (shrink)

The objective of Working Group 4 of the COST Action NET4Age-Friendly is to examine existing policies, advocacy, and funding opportunities and to build up relations with policy makers and funding organisations. Also, to synthesize and improve existing knowledge and models to develop from effective business and evaluation models, as well as to guarantee quality and education, proper dissemination and ensure the future of the Action. The Working Group further aims to enable capacity building to improve interdisciplinary participation, to promote knowledge (...) exchange and to foster a cross-European interdisciplinary research capacity, to improve cooperation and co-creation with cross-sectors stakeholders and to introduce and educate students SHAFE implementation and sustainability. To enable the achievement of the objectives of Working Group 4, the Leader of the Working Group, the Chair and Vice-Chair, in close cooperation with the Science Communication Coordinator, developed a template to map the current state of SHAFE policies, funding opportunities and networking in the COST member countries of the Action. On invitation, the Working Group lead received contributions from 37 countries, in a total of 85 Action members. The contributions provide an overview of the diversity of SHAFE policies and opportunities in Europe and beyond. These were not edited or revised and are a result of the main areas of expertise and knowledge of the contributors; thus, gaps in areas or content are possible and these shall be further explored in the following works and reports of this WG. But this preliminary mapping is of huge importance to proceed with the WG activities. In the following chapters, an introduction on the need of SHAFE policies is presented, followed by a summary of the main approaches to be pursued for the next period of work. The deliverable finishes with the opportunities of capacity building, networking and funding that will be relevant to undertake within the frame of Working Group 4 and the total COST Action. Relative Energy Deficiency in Sport (RED-S) is a syndrome of impaired health and performance that occurs as a result of low energy availability (LEA). Whilst many health effects associated with RED-S have been widely studied from a physiological perspective, further research exploring the psychological antecedents and consequences of the syndrome is required. Therefore, the aim of this study was to qualitatively explore athlete experiences of RED-S. Twelve endurance athletes (female n= 10, male n= 2; M age = 28.33 years) reporting (...) past or current experiences of RED-S, associated with periods of LEA, took part in semi-structured interviews designed to explore: contexts and mechanisms underpinning the onset of RED-S; the subjective experience of RED-S; and contexts and mechanisms influencing “recovery” from RED-S. Regardless of how RED-S was initiated, all athletes experienced a multitude of physiological impairments, accompanied by significant psychological distress. This paper contributes novel understanding of the complex interplay between physiological and psychological components of RED-S from the perspective of information-rich cases. The findings suggest that system-wide educational prevention and awareness interventions are vital for athletes and support personnel, such as coaches, parents, dieticians, psychologists, and sports medicine staff. In ‘Colonialism, territory and pre-existing obligations,’ Cara Nine argues that Lea Ypi’s account of the wrongness of colonialism has a hole in it: Ypi leaves open the possibility of justified settler colonialism. Nine suggests that we can patch this hole by attaching value to existing political associations. But Nine’s solution has its own hole. Many political associations exist due to settler colonialism, and thus if we endorse the value of these associations we seem to endorse colonialism. In response, we could (...) say that past colonial injustices have been remediated or superseded. Or, we could leave the hole open: colonialism is sometimes justified. I argue that, surprisingly, the second option is likely preferable, because it does not require us to say that the injustices of colonialism are gone, and because it points towards a better theory of colonialism’s wrongness than Ypi’s: cosmopolitan instrumentalism, according to which colonialism is wrong only when it is unjust according to one’s overall theory of cosmopolitan justice. In recent work, Andrew T. Forcehimes and Robert B. Talisse correctly note that G.A. Cohen’s fact-insensitivity thesis, properly understood, is explanatory. This observation raises an important concern. If fact-insensitive principles are explanatory, then what role can they play in normative deliberations? The purpose of my paper is, in part, to address this question. Following David Miller, I indicate that on a charitable understanding of Cohen’s thesis, an explanatory principle explains a justificatory fact by completing an otherwise logically incomplete inference. As (...) a result, the explanatory role such a principle plays is inseparable from its status as a (not necessarily successful) justificatory reason. With this interpretation in hand, I then proceed to argue that Lea Ypi’s and Robert Jubb’s recent criticisms fail to undermine Cohen’s thesis, and that fact-insensitive principles, once discovered, are especially helpful for purposes of deliberation in circumstances characterized by changing and changeable feasibility constraints. One is the Lack-of-evidence Argument (LEA), according to which there would have been good objective evidence of God's existence if he were to exist. The other is the Argument from Nonbelief (ANB), according to which there would not be as many nonbelievers as there actually are if God were to exist. Cunoașterea și informațiile (abordate în ansamblu sau în componentele lor distincte) sunt o preocupare majoră pentru tehnologia informației, sisteme de informații, știința informației și activitatea de informații în general. Procesul obţinerii, prelucrării şi analizei informaţiilor este o preocupare majoră pentru societatea actuală. În acest scop se folosesc procedee şi tehnici specifice pentru culegerea sau generarea de informaţii, prelucrarea acestora prin analiză şi sinteză, generarea de predicţii şi strategii, transmisia şi prezentarea informaţiilor factorilor de decizie, şi stocarea lor. Analiza informațiilor poate (...) fi caracterizată ca o intreprindere epistemologică, care urmărește să dezvolte o înțelegere clară a produselor cunoașterii. Perspectiva lui Polanyi despre cunoaștere servește unor considerații epistemologice fundamentale privind cunoașterea ca produs. Conceptul acesta de cunoaștere personală contribuie la o înțelegere mai profundă a cunoașterii. Perspectiva inversează ierarhia tradițională epistemologică. Cea mai mare provocare ar putea fi aceea de a convinge analiștii, conducerea și clienții lor de avantajele metodelor analitice structurale. O introducere treptată și naturală a acestor metode în fluxul obișnuit de procese și produse de informații ar putea fi mai eficientă decât realinierea la scară largă a gândirii. "Spațiul" activităților de informații s-a extins de la guverne ca actori statali la actori non-statali precum ONG-uri, instituții transnaționale și corporații, firme private și grupuri de interese și presiuni reprezentând comunități, concepte și ideologii diverse. Procesul de luare a deciziilor a devenit dispersat și granular. Actuala "democratizare" a serviciilor de informații are un impact major asupra colectării și analizei informațiilor, și a procesului decizional, determinând serviciile de informații să devină mai transparente, să ia în considerație dezbaterile publice a acțiunilor lor și să fie nevoite, uneori, să le justifice public. Un domeniu de care pot beneficia serviciile de informații este cel al managementului cunoașterii, implicat într-o dezbatere riguroasă asupra conceptelor, teoriilor și abordărilor cunoașterii și utilizării acestora. RESUMO A fêmea bovina apresenta ciclos estrais em intervalos de 19 a 23 dias e estes só são interrompidos durante a gestação ou devido (...) a alguma patologia. O estro é o período de aceitação da cópula e tem uma duração de 8 a 18 horas. Durante o metaestro ocorre a ovulação e se desenvolve o corpo lúteo. O diestro é o estágio mais longo do ciclo e é caracterizado pela presença de um corpo lúteo. Se a gestação não for estabelecida, o endométrio segrega prostaglandina F2α(PGF2α) o que induz a luteólise, reiniciando assim um novo ciclo. 2. EIXO HIPOTÁLAMO-HIPÓFISE-OVÁRIO As hormonas são substâncias produzidas por diferentes células do organismo que exercem funções específicas em outras células (células brancas). Algumas hormonas atuam na mesma célula que a secreta (atividade autocrina), outras nas células vizinhas (atividade parácrina) e outras são transportadas pelo sangue e exercem a sua função em células de outros órgãos (atividade endócrina). Existem outros tipos de hormônios que comunicam a diferentes indivíduos e são conhecidos como feromônios. Os feromônios regulam diferentes funções, entre as quais se destacam as reprodutivas. O hipotálamo encontra-se na base do cérebro, é formado por núcleos pares de neurônios e comunica-se com a hipófise através de um sistema circulatório especializado conhecido como sistema porta-hipotálamo-hipofisário. Os neurônios da área ventromedial e da área pré-óptica do hipotálamo secretam a hormona libertadora das gonadotropinas (GnRH), que por sua vez chega à hipófise através do sistema porta-hipotálamo-hipofisário e estimula a secreção da hormona luteinizante (LH) e da hormona folículo estimulante (FSH). A LH mantém um padrão de secreção paralelo à secreção da GnRH; ou seja, uma parcela de GnRH corresponde a uma parcela de LH, ao contrário da FSH que tem uma produção basal elevada inibida pelo estradiol e inibina, por este motivo, a sua secreção não apresenta um padrão pulsante semelhante à LH. A GnRH tem duas formas de secreção: a primeira é pulsante ou tônica, regulada por estímulos externos (fotoperíodo, bioestimulação, amamentação) e por estímulos internos (metabolitos, hormonas metabólicas, hormonas sexuais); a segunda forma é pré-ovulatória ou cíclica e é estimulada pelos estrogênios durante o estro e inibida pela progesterona. A secreção de alguns hormônios, bem como diversos processos fisiológicos, são sincronizados com a duração do dia e da noite (ritmos endógenos). A luz é percebida pelos fotorreceptores da retina e o sinal luminoso chega à glândula pineal através de conexões neuronais (trato retino-hipotalâmico). Na glândula pineal, o estímulo produzido pela luz inibe a síntese da melatonina. Desta forma, a duração do dia e da noite (fotoperíodo) é registada pelas variações nas concentrações da melatonina. Na vaca, sabe-se que o fotoperíodo influencia alguns processos reprodutivos, embora não seja, em sentido estrito, uma espécie com um padrão reprodutivo sazonal. Os feromônios sexuais são excretados através da urina, fezes e fluidos corporais; eles são percebidos pelo epitélio olfatório e órgão vomeronasal. Posteriormente, algumas vias nervosas, estimulam no hipotálamo a frequência dos pulsos de secreção da GnRH. A exposição a feromonas femininas provoca no macho um aumento na frequência de secreção do LH e isto por sua vez aumenta as concentrações de testosterona. Os feromônios masculinos induzem na fêmea um aumento da frequência de secreção do LH, estimulando o crescimento folicular e a secreção de estradiol. A estimulação sexual provocada pelo macho ou pela fêmea é denominada bioestimulação. As alterações na condição corporal estão positivamente correlacionadas com as concentrações séricas de insulina, fator de crescimento semelhante à insulina tipo I (IGF- I) e leptina. Assim, quanto maior a classificação da condição corporal, maior é a concentração sérica destas hormonas, que atuam como sinais que chegam ao hipotálamo e modificam a frequência de secreção da GnRH. Por exemplo, a transição do anestro para a ciclicidade coincide com um aumento da condição corporal e das concentrações de insulina, IGF- I e leptina (figura 1). Figura 1. Os estrogênios podem ter um feedback positivo ou negativo sobre a secreção da GnRH, o que depende da fase do ciclo reprodutivo. Em animais pré-púberes e em anestro pós-parto, os estrogênios inibem a secreção de GnRH, mas durante o período de proestro e estro há uma estimulação para a secreção de GnRH. A progesterona reduz a secreção da GnRH, bem como a resposta da hipófise à GnRH, inibindo assim a maturação folicular e a ovulação. Por esta razão, a progesterona foi utilizada com sucesso como contraceptivo em humanos e para o controle artificial da reprodução em animais domésticos (figura 2). Figura 2. Na fase pré-púbere e no anestro pós-parto, os estrogênios inibem a secreção de GnRH, enquanto no proestro e estro, estimulam-na. A progesterona inibe a secreção da GnRH e diminui a resposta da hipófise à GnRH. Os estrogênios e a inibina suprimem a secreção de FSH diretamente na hipófise. Fonte: GALINA, et al. 2008. Os neurônios secretores da GnRH não têm receptores para estrogênios nem progesterona, pelo que estas hormonas não têm forma de regular diretamente a secreção da GnRH. Existe um grupo de neurônios hipotalâmicos que exprimem o gene Kiss-1 que codifica o peptídeo kisspeptina. Os neurônios secretores da GnRH têm receptores para este peptídeo, de modo que a kisspeptina fornece a informação aos neurônios secretores da GnRH em relação às concentrações de hormônios sexuais. A kisspeptina é um potente estimulador (secretagogo) da secreção da GnRH e é muito provável que nos próximos anos venha a fazer parte dos recursos hormonais para o controle artificial da reprodução, não só nos bovinos, mas em todas as espécies domésticas. 3. DESENVOLVIMENTO FOLICULAR O ovário é responsável pela produção de ovócitos e pela síntese de hormônios sexuais, estrogênios e progesterona, que promovem e regulam a fertilização do ovócito e a manutenção da gestação. O ovócito encontra-se no interior do folículo ovárico rodeado por células granulosas que participam de forma ativa no seu crescimento e maturação. As experiências in vitro demonstram a dependência dos ovócitos das células da granulosa, assim, quando os ovócitos são induzidos a amadurecer devem estar rodeados por várias camadas de células da granulosa para que este processo seja bem sucedido, caso contrário, não adquirem o potencial para desenvolver um embrião. Embora as células da teca interna não estejam em contato direto com o ovócito, seu papel na maturação deste o exercem mediante a produção de andrógenos, mesmos que são convertidos em estrogênios pelas células da granulosa. Além disso, as células da teca favorecem o estabelecimento da rede capilar que apoia o desenvolvimento folicular. Por outro lado, os novos conhecimentos indicam que o ovócito não é um elemento passivo no desenvolvimento folicular, mas regula a função das células foliculares; o que significa que ele próprio participa na criação de um microambiente ideal para a sua maturação. Além disso, é possível que o ovócito tenha um papel na ativação do desenvolvimento dos folículos primordiais. A fêmea bovina nasce com aproximadamente 200 mil folículos, dos quais muito poucos se ativam e iniciam seu crescimento, e a maior parte deles sofre atresia em diferentes etapas de desenvolvimento. Ao nascimento, os folículos estão na fase mais elementar e são conhecidos como folículos primordiais. Posteriormente estes folículos se ativam e se transformam em folículos primários e secundários; até este momento os folículos não têm antro (etapa pré-antral) e seu desenvolvimento é independente das gonadotropinas. Quando os folículos formam o antro são conhecidos como folículos terciários e seu desenvolvimento é dependente das gonadotropinas (etapa antral). O crescimento folicular no estágio antral ocorre em forma de ondas e cada onda começa com um aumento nos níveis de FSH, o qual promove o crescimento de um grupo de cinco a seis folículos (~4 mm de diâmetro); este processo é conhecido como recrutamento. Subsequentemente, um único folículo continua a crescer (folículo dominante), o que provoca um aumento das concentrações de estrogênios e inibina, uma diminuição das concentrações de FSH e atresia dos folículos subordinados, pois eles dependem totalmente desta hormona, enquanto o folículo dominante continua o seu desenvolvimento estimulado pela LH. O folículo dominante perdura de quatro a seis dias e se não chega a ovular, sofre atresia. Após a atresia do folículo dominante, diminuem-se os níveis de estrogênio e inibina, observa-se um aumento das concentrações de FSH e inicia-se uma nova onda folicular. O folículo dominante que está presente quando o corpo lúteo sofre regressão, continua seu desenvolvimento e ovula, em resposta ao pico pré-ovulatório de LH. Além de promover a liberação do ovócito, a secreção pré-ovulatória de LH regula a formação do corpo lúteo a partir das células foliculares, processo conhecido como luteinização. Durante o ciclo estral são apresentadas de duas a três ondas foliculares. As vacas com três ondas foliculares têm uma fase lútea mais longa e, consequentemente, um ciclo estral mais longo, de 22 a 23 dias; enquanto as vacas com duas ondas apresentam um ciclo estral de 18 a 21 dias. Nas vacas leiteiras, cerca de 70% apresentam duas ondas foliculares, enquanto 30% exibem três ondas (figura 3 e 4). Nas vacas com duas ondas foliculares, o período de dominação folicular é maior do que nas de três ondas. O tempo de dominação influencia o potencial dos ovócitos para desenvolver um embrião viável; assim, a porcentagem de concepção é menor quando ovulam folículos que tiveram mais dias de dominação dos que quando ovulam folículos com menor tempo de dominação (figura 5). Cada onda começa com um aumento nas concentrações de FSH, o que promove o recrutamento de cinco a seis folículos (~4 mm de diâmetro). Posteriormente um único folículo continua crescendo (folículo dominante), enquanto seus companheiros (subordinados) sofrem atresia. O folículo dominante perdura de quatro a seis dias e se não chega a ovular, sofre atresia. Após a atresia do folículo dominante observa-se um aumento das concentrações de FSH, iniciando-se uma nova onda folicular. 3.1 Ovulação múltipla Nos últimos anos tem-se observado um aumento na proporção de vacas com ovulação múltipla (20% x 1% em novilhas), o que tem provocado um aumento da proporção de partos gêmeos (8% x 1% em novilhas). A frequência de vacas com ovulação múltipla está associada com a alta produção de leite; de modo que, as vacas que produzem menos de 40 kg mostram 6% de ovulações múltiplas e aquelas que produzem mais de 50 kg alcançam até 50%. A causa deste fenômeno ainda é obscura, contudo, observaram-se diferenças nas concentrações de FSH, de tal forma que as vacas que desenvolvem de dois a três folículos dominantes numa onda folicular, apresentam níveis de FSH mais elevados que as vacas que têm apenas um folículo dominante. Em vacas em lactação, a concentração de progesterona é baixa devido ao aumento do metabolismo hepático, o que aumenta a sua taxa de eliminação. Foi observado que as vacas que tiveram uma fase lútea com níveis de progesterona mais elevados, no ciclo anterior à inseminação, apresentam menos ovulações múltiplas em comparação com as vacas que tiveram níveis de progesterona mais baixos. Propõe-se que as baixas concentrações de progesterona permitam um aumento da frequência de secreção da GnRH e, consequentemente, da LH e da FSH, favorecendo a predominância múltipla e, eventualmente, a ovulação de mais de um folículo. Nos rebanhos leiteiros, as gestações gêmeas não são desejáveis porque aumenta o risco de perda da gestação e, se esta chegar ao término, haverá o risco de perda da gestação e, se esta for concluída, o risco de distorcia é consideravelmente mais elevado (figuras 6, 7, 8 e 9). DESENVOLVIMENTO E CONTROLE DA FUNÇÃO DO CORPO LÚTEO Quando o folículo dominante completa sua maturação, ele produz níveis de estrogênio suficientes para provocar a liberação máxima da GnRH, o que desencadeia o pico pré-ovulatório da LH. Esta secreção de LH provoca a ovulação e inicia as mudanças para que o folículo se transforme em um corpo lúteo, processo conhecido como luteinização. A luteinização compreende todas as mudanças morfológicas, endócrinas e enzimáticas que ocorrem no folículo ovulatório até que este se transforme num corpo lúteo. O processo de luteinização começa a partir da elevação pré-ovulatória de LH; mesmo antes da ovulação. A luteinização do folículo dominante (≥8 mm de diâmetro) pode ser induzida hormonalmente pela injeção de GnRH ou gonadotropina coriónica humana (hCG). A ovulação ocorre em média 30 horas após o pico pré-ovulatório de LH. A secreção pré-ovulatória de LH desencadeia a liberação de enzimas proteolíticas e de mediadores da inflamação na parede folicular, as quais degradam o tecido conjuntivo e ocasionam morte celular. Posteriormente, a PGF2α induz contrações da teca externa, levando à ruptura folicular e à expulsão do ovócito. Após a ovulação, as células da teca interna e da granulosa migram e distribuem-se nas paredes do folículo. As células da teca interna se diferenciam e se multiplicam em células lúteas pequenas, enquanto que as células da granulosa se hipertrofiam e dão origem às células lúteas grandes. Estas alterações são facilitadas pela ruptura da membrana basal que separa a camada celular da granulosa da teca interna. Em forma paralela começa a formação de uma ampla rede de capilares que se distribuem em todo o corpo lúteo em formação, e chegam a constituir até 20% do volume desta estrutura (figuras 10 e 11). A progesterona é o principal produto de secreção do corpo lúteo. No quinto dia do ciclo estral, as concentrações séricas desta hormona são superiores a 1 ng/ ml, indicando que o corpo lúteo adquiriu a sua plena funcionalidade. A progesterona atua basicamente sobre os órgãos genitais da fêmea, sendo responsável pela preparação do útero para o estabelecimento e manutenção da gestação. Na mucosa do oviduto e do útero, estimula a secreção de substâncias que promovem o desenvolvimento do embrião, até que este comece a nutrir-se através da placenta. A progesterona suprime a resposta imunitária do útero, o que é necessário para tolerar o embrião, já que este é um tecido estranho para a vaca. Além disso, a progesterona evita as contrações do útero, fecha o colo do útero e modifica as características do muco cervical, tornando-o mais viscoso, impedindo a passagem de agentes estranhos para o interior do útero. Na glândula mamária estimula o desenvolvimento do sistema alveolar, preparando-a para a síntese e a secreção de leite. 5. REGRESSÃO DO CORPO LÚTEO A regressão lútea é um processo ativo ocasionado pela secreção uterina da PGF2α. O mecanismo pelo qual se inicia a síntese e secreção da PGF2α depende de uma interação entre o corpo lúteo, os folículos e o útero. Os estrogênios produzidos no folículo dominante desempenham um papel importante no início da secreção de PGF2α, uma vez que promovem a síntese de receptores para oxitocina. Além disso, os estrogénios estimulam no endométrio a produção da fosfolipase A e da ciclooxigenase; enzimas indispensáveis para a síntese da PGF2α. Durante o ciclo estral, a progesterona inibe a síntese da PGF2α através da supressão da formação de receptores para o estradiol. Após um período de 12 a 14 dias de exposição à progesterona, as células endometriais tornam-se insensíveis à progesterona. Quando isso ocorre, as células endometriais sintetizam receptores para estradiol, permitindo que o estradiol produzido no folículo dominante estimule a síntese de receptores para oxitocina. Neste momento, o endométrio está pronto para sintetizar e secretar PGF2α, em resposta ao estímulo da oxitocina. A primeira secreção de oxitocina é de origem hipotalâmica, o que desencadeia o primeiro pulso de PGF2α. Os seguintes episódios de PGF2α são induzidos pela oxitocina produzida no corpo lúteo. A PGF2α é secretada em episódios (pulsos) com intervalos de seis a oito horas, sendo necessários cinco a seis episódios para a luteólise ocorrer. Se a PGF2α não seguir este padrão de secreção, a regressão do corpo lúteo falhará. Além da PGF2α de origem uterina, o corpo lúteo também produz PGF2α, que aumenta o efeito luteolítico. A falta de sensibilidade à PGF2α observada nos corpos lúteos imaturos (primeiros cinco dias após a ovulação) deve-se ao fato de, neste período, o corpo lúteo ainda não produzir PGF2α (figura 12, 13 e 14). 6. ETAPAS DO CICLO ESTRAL O ciclo estral é dividido em quatro etapas bem definidas. 6.1 Estro Neste estágio a fêmea aceita a cópula ou a monta de outra vaca. O estro é provocado pelo aumento significativo das concentrações de estradiol produzido pelo folículo pré-ovulatório e pela ausência de um corpo lúteo. A duração desta etapa é de 8 a 18 horas. 6.2 Metaestro O metaestro é a etapa posterior ao estro, tem uma duração de quatro a cinco dias. Durante esta etapa ocorre a ovulação e se desenvolve o corpo de lúteo. Após a ovulação, observa-se uma depressão no lugar ocupado pelo folículo ovulatório (depressão ovulatória) e, posteriormente, se desenvolve o corpo hemorrágico (corpo lúteo em processo de formação). Durante o metaestro, as concentrações de progesterona começam a aumentar até atingirem níveis superiores a 1 ng/ml, momento a partir da qual considera-se que o corpo lúteo atingiu a maturidade. O momento em que as concentrações de progesterona são superiores a 1 ng/ml toma-se como critério fisiológico a determinação do fim do metaestro e o início do diestro. Um evento hormonal que se destaca neste período consiste na apresentação do pico pós-ovulatório de FSH, o qual desencadeia a primeira onda de desenvolvimento folicular. Algumas vacas apresentam sangramento conhecido como sangramento metaestral (figura 15). 6.3 Diestro O diestro é o estágio de maior duração do ciclo estral, de 12 a 14 dias. Durante este estágio o corpo lúteo mantém sua plena funcionalidade, o que se reflete em concentrações sanguíneas de progesterona, maiores que 1 ng/ml. Além disso, nesta fase, pode-se encontrar folículos de tamanho diferente devido às ondas foliculares. Após 12-14 dias de exposição à progesterona, o endométrio começa a secretar PGF2α em um padrão pulsátil, ao qual termina com a vida do corpo lúteo e com o diestro. Em termos endócrinos, quando o corpo lúteo perde a sua funcionalidade, ou seja, quando as concentrações de progesterona diminuem abaixo de 1 ng/ml, finaliza-se o diestro e começa o proestro. Convém mencionar que durante esta fase, a LH é secretada com uma frequência muito baixa e a FSH tem incrementos responsáveis pelas ondas foliculares. 6.4 Proestro O proestro caracteriza-se pela ausência de um corpo lúteo funcional e pelo desenvolvimento e maturação do folículo ovulatório. O proestro na vaca dura de dois a três dias. Um evento hormonal característico desta etapa é o aumento da frequência dos pulsos de secreção de LH que levam à maturação final do folículo ovulatório e ao aumento do estradiol sérico, que desencadeia o estro. Para além da classificação do ciclo estral acima descrita, existe outra que divide o ciclo em duas fases: progestacional (lútea) e estrogênica (folicular). CONCLUSÕES PRÉVIAS  O ciclo estral dura de 19 a 23 dias.  A vaca é receptiva durante 8 a 18 horas (estro).  Ao nascimento uma bezerra tem cerca de 200 mil folículos primordiais.  Durante o ciclo estral se apresentam de duas a três ondas foliculares.  De cinco a seis folículos são recrutados em cada onda folicular.  Cerca de 70% das vacas têm duas ondas foliculares e 30% apresentam três ondas.  Entre 10 e 20% das vacas têm ovulações múltiplas (dois a três folículos) e 8% têm partos gêmeos.  A ovulação ocorre 30 horas após o pico pré-ovulatório de LH.  A secreção pré-ovulatória de LH é de 15 a 30 ng/ml.  12 a 14 dias são necessários para que o endométrio se torne insensível à progesterona e comece a secretar PGF2α. - São necessários cinco a seis pulsos de PGF2α com um intervalo de oito horas para ocasionar a luteólise.  O corpo lúteo não é sensível à PGF2α nos primeiros cinco dias do ciclo estral. DESENVOLVIMENTO EMBRIONÁRIO E DIFERENCIAÇÃO SEXUAL -/- E. I. C. da Silva Departamento de Agropecuária – IFPE Campus Belo Jardim Departamento de Zootecnia – UFRPE sede -/- 1.1 INTRODUÇÃO O sexo foi definido como a soma das diferenças morfológicas, fisiológicas e psicológicas que distinguem o macho da fêmea permitindo a reprodução sexual e assegurando a continuidade das espécies. Os processos de diferenciação sexual são realizados durante o desenvolvimento embrionário, onde ocorre a proliferação, diferenciação e maturação das células germinativas e primordiais, precursoras (...) de ovócitos e espermatozoides em fêmeas e machos, respectivamente. Assim, os embriões machos e fêmeas iniciam o seu desenvolvimento de forma semelhante, de modo que em ambos os sexos se estabelecem em estruturas idênticas a partir das quais se formarão os órgãos reprodutores correspondentes a cada sexo. O conhecimento da origem e do desenvolvimento do aparelho genital é indispensável para entender sua função e as alterações que produzem infertilidade ou esterilidade. 1.2 DETERMINAÇÃO DO SEXO CROMOSSÔMICO Nos mamíferos, o sexo cromossômico é determinado no momento da fertilização, quando um óvulo, que contém um cromossomo X, é fecundado por um espermatozoide portador de um cromossomo X ou um cromossomo Y. No primeiro caso, o complemento cromossômico seria XX, o que originaria uma fêmea (sexo homogamético), e no segundo daria como resultado um macho com a fórmula cromossômica XY (sexo heterogâmico). 1.3 A GÔNADA INDIFERENCIADA A primeira manifestação das gônadas se aprecia no embrião em forma de um par de eminências longitudinais chamadas cristas ou dobras gonodais, situadas em ambos os lados da linha média entre os mesonefros (rins em desenvolvimento) e do mesentério dorsal. Nos embriões dos mamíferos, as células germinativas primordiais (CGP) manifestam-se em estágios precoces do desenvolvimento, podendo ser detectadas pela primeira vez na metade da gastrulação. As CGP são células grandes, de citoplasma claro e núcleo grande e redondo, localizadas na parede do saco vitelino, perto do alantoide. Essas células possuem grande capacidade de proliferação e vão migrar desde o endoderma do intestino e o epitélio do saco vitelino, através do mesentério, até as cristas gonodais. Isso ocorre por volta do 26° dia da gestação no bovino. Sua migração realiza-se graças aos movimentos de translocação passiva e deslocamento ameboide ativo. Desconhece-se o mecanismo pelo qual estas células são dirigidas para as cristas gonodais, porém foram estudadas algumas moléculas que se expressam durante sua migração e que poderiam desempenhar um papel importante na diferenciação deste tipo celular. A fosfatase alcalina é uma enzima que tem sido usada como marcador de CGP para determinar a sua origem e migração. Num estudo recente, foi inserido um marcador fluorescente que se exprime unicamente nas células germinativas primordiais de embriões transgênicos, e utilizando este marcador e a fosfatase alcalina determinou-se a origem e o padrão de migração destas células. O primeiro sinal de diferenciação das células germinativas primordiais é a expressão de fosfatase alcalina, e esta apareceu pela primeira vez na parte mais posterior da linha primitiva. No sétimo dia de desenvolvimento no embrião do camundongo, o endoderma visceral (AF+) é substituído pelo endoderma definitivo (AF-) originado na parte anterior da linha primitiva. O fator de transcrição Oct-4 é expresso nas CGP de ambos os sexos, pelo que acredita-se estar envolvido na mantença a totipotêncialidade das células. O receptor tirosina quinase, cujo ligante é o fator de Steel, é outra das moléculas que expressam as CGP. Tem sido demonstrado que este receptor possui um papel muito importante na sobrevivência deste tipo celular. Existem outros fatores que promovem a sobrevivência e/ou proliferação de CGP in vitro. Em experiências realizadas com o fator de transformação beta I (TGFβ-I), observou-se que este tem um efeito negativo sobre a proliferação das CGP. Outra atividade que tem sido postulada a este fator é o de um agente quimioatraente que possivelmente possa direcionar a migração destas células para a gônada. a) Um formado pelas células germinativas primordiais (precursoras dos gametas masculinos ou femininos), rodeadas de células somáticas das quais posteriormente se derivarão as células de Sertoli no macho e as células da granulosa na fêmea. b) O tecido que formará o estroma da gônada: tecido conjuntivo, vasos sanguíneos e as células intersticiais com atividade esteroidogênica (células de Leydig no testículo e a teca interna do ovário). As células somáticas do primórdio gonodal originam-se do mesoderma. Inicialmente são de três tipos: mesenquimáticas, mesoteliais e endoteliais. As células mesenquimáticas e mesoteliais iniciam grande atividade proliferativa ao chegar as CGP. Observa-se então uma condensação de células de origem mesotelial e mesenquimatoso que forma um agregado compacto denominado "blastema gonodal". A partir deste primórdio embrionário, diferenciam-se dois tecidos gonodais: os cordões sexuais e o estroma. Os cordões sexuais são arranjos epiteliais que se encontram delimitados por uma folha basal, e dentro deles encontramos as CGP. Por sua vez, no estroma encontram-se células do tipo mesenquimático e vasos sanguíneos. Neste momento, as gônadas são indiferenciadas e bipotencialmente sexuais, sendo impossível diferenciar, morfologicamente, uma gônada masculina de uma feminina, mas no caso dos machos genéticos já existe uma diferenciação da gônada a nível molecular. Nesta fase já se encontram presentes as estruturas das quais se desenvolvem os dutos mesonéfricos ou de Wolff precursores do aparelho genital masculino e os dutos paramesonéfricos ou de Müller que darão origem ao aparelho reprodutor feminino. Há uma série de fatores envolvidos no desenvolvimento precoce da gônada, entre os quais o fator esteroidogênico I (SFI: Steroidogenic fator l), que é um membro da subfamília de receptores nucleares, receptores órfãos. Este fator de transcrição tem um local de ligação ao DNA composto por dois dedos-de-Zinc. O SFI foi identificado como um ativador de genes envolvidos na biossíntese de esteroides em diferentes células. O SFI está presente durante o desenvolvimento embrionário em regiões associadas com funções endócrinas como gônadas, adrenais, pituitárias e hipotálamos. Os animais homozigotos para o gene SFI defeituoso, necessitam de gônadas e adrenais e têm a função gonadotrófica alterada. Os ratos sem SFI carecem de gonadotrofos e têm um desenvolvimento anormal do núcleo ventro-medial do hipotálamo; em particular as gônadas deixam de se desenvolver entre os dias 11 a 15 e degeneram-se por apoptose. No entanto, a crista genital forma-se e é colonizada pelas células germinativas, o que indica que estas continuam a receber o sinal adequado para a sua migração. Portanto, o SFI não está envolvido no desenvolvimento precoce da gônada e do sistema urogenital, mas parece estar envolvido na manutenção do crescimento das células somáticas presentes na gônada indiferenciada. O gene associado ao tumor de Wilms (WTI: Wilm's tumor Associated) está envolvido no desenvolvimento da gônada e do rim. Durante o desenvolvimento embrionário, WTI se expressa em todo o mesoderma intermediário e posteriormente na gônada indiferenciada, bem como no rim em formação. WTI regula o sinal indutivo do mesênquima para o epitélio celômico dos mesonefros. Se este for o caso, então WTI é responsável pelo crescimento da crista genital ao dirigir a entrada do epitélio celômico. Dado que estas células darão origem às células de Sertoli, a carência de WTI pode causar o desenvolvimento de embriões XY como fêmeas simplesmente porque não se formam as células de Sertoli. Em geral, todos os genes importantes na diferenciação do mesoderma intermediário e do sistema urogenital intervêm no desenvolvimento da gônada precoce. 1.4 DIFERENCIAÇÃO GONODAL O desenvolvimento das gônadas e ductos genitais descritos até o momento, é o mesmo para ambos os sexos. Igualmente, os genes descritos, que estão envolvidos no desenvolvimento das gônadas, ductos genitais e migração das células germinativas, afetam igualmente os embriões com genótipo XX ou XY. A gônada primitiva consiste anatomicamente de uma medula (interna) e uma crosta (externa), e de acordo com o local onde ocorre a colonização das células germinativas, diferenciara em testículo ou um ovário, respectivamente. Nos mamíferos, a primeira manifestação estrutural de diferenciação sexual é detectada na gônada dos machos, onde as células germinativas estão localizadas na medula. A diferenciação do testículo inicia-se quando os cordões sexuais se separam do epitélio celômico como consequência dos arranjos produzidos por uma invasão do mesênquima e vasos sanguíneos que provoca a compactação dos cordões, agora denominados cordões testiculares. As células que rodeiam os cordões se achatam e formam as células mioides, que são responsáveis pela formação das membranas basais. As células do epitélio interno, ou seja, as células de Sertoli, têm duas funções principais: o suporte das CGP e a síntese da hormona antimulleriana, responsável pela regressão dos ductos de Müller e secretada durante o período de diferenciação sexual. As células do estroma que rodeiam os cordões testiculares diferenciam-se para formar vários tipos de células: células mioides, fibroblastos, endotélio e células de Leydig, que são as mais importantes pela sua atividade endócrina. Posteriormente, os cordões testiculares dão origem aos túbulos seminíferos, que contêm o epitélio que produzirá os espermatozoides ao chegar à puberdade. Na fêmea, durante os estágios iniciais de diferenciação gonodal, não se observam mudanças em relação à gônada indiferenciada, só pode-se observar um certo crescimento devido à proliferação de células somáticas e germinativas. As células germinativas iniciam um período de proliferação, que termina com o início da meiose. Iniciada a meiose, dá-se o processo de foliculogênese; neste momento os cordões epiteliais se fragmentam, de tal maneira que cada ovócito fica rodeado de células epiteliais cobertas por uma folha basal fina (figura 1). Para que a gônada primitiva se desenvolva em testículo é indispensável a presença do cromossoma Y, independentemente do número de cromossomas X que contenha o genoma de um indivíduo. O gene determinante do testículo encontra-se localizado no cromossoma Y, denominado sry em ratos e SRY em humanos. O gene sry se expressa durante o desenvolvimento embrionário na crista genital de embriões de camundongos. A expressão é detectável no dia 10,5, pouco depois do aparecimento das cristas genitais, atinge o seu máximo durante o dia 11,5 e mantém-se até pouco depois de ocorrerem os primeiros sinais morfológicos de diferenciação testicular no dia 12,5. Este padrão de expressão é compatível com a teoria de que sry atua induzindo a ativação dos genes (figura 2) que conduzem ao desenvolvimento testicular, sem que exista a necessidade da expressão contínua de sry para manter a diferenciação do testículo após o dia 12,5. Como mencionado anteriormente, a gônada primitiva é composta por vários tipos de células. No entanto, as células germinativas primordiais não são o local de expressão do sry, já que os embriões que necessitam de células germinativas mantêm a expressão de sry e desenvolvem o sexo gonodal normalmente. As células somáticas na gônada em desenvolvimento incluem também as células de suporte. Sabe-se que é nestas células que o sry é expresso para que se diferenciem em células de Sertoli, e a expressão transitória de sry indica que deve ativar a outros genes para a manutenção das células de Sertoli. Uma vez diferenciadas as células de Sertoli, elas se encarregarão da diferenciação do resto das células na gônada. -/- Figura 1: Representação da diferenciação dos órgãos genitais internos. Embora não se conheçam os genes que provavelmente regulam esse gene, estudos realizados em camundongos demonstram que este gene parece coordenar-se com certos genes autossômicos. Entre estes genes autossômicos, o sox9, que é produzido pelas células de Sertoli uma vez que são estimuladas por sry, de modo que sox9 é um dos genes relacionados estruturalmente com sry. O Sox9 funciona como um fator de transcrição, mas não se sabe se a proteína tem qualquer outra função estrutural; este gene exprime-se abundantemente nos condrócitos e está relacionado com defeitos do aparelho ósseo chamados displasia campomélica. Curiosamente, os pacientes XY com esta condição sofrem frequentemente de reversão do sexo. O Sox9 é um dos poucos genes, além do SRY, do qual as mutações demonstraram interferir com a determinação sexual masculina. No entanto, apenas 75% dos pacientes com anomalias esqueléticas de tipo displasia campomélica têm reversão sexual e não foram encontrados casos de reversão sexual devido a um defeito de Sox9 que não seja acompanhado de defeitos esqueléticos. Isso indica que o Sox9 é apenas um membro da rede de genes que são ativados para determinar a diferenciação sexual, enquanto a rota que rege a condrogênese é mais sensível a perturbações deste. O momento em que se detecta a expressão do gene Sox9 (11dpc em ratos) coincide com a máxima expressão de sry, o que poderia indicar a possibilidade de que sry regule positivamente a Sox9. De fato, na região do promotor de Sox9 há um local de união ao que potencialmente se pode unir o sry. A expressão de Sox9 durante a diferenciação sexual sugere um papel abaixo de sry na diferenciação das células de Sertoli. O cromossoma X também é importante na diferenciação gonodal. O gene DAX-I foi isolado do lócus DSS (Dosage sensitive sex reversal) do cromossoma X. DAX-I é parte da cascata de determinação sexual, mas não é necessário para a formação do testículo. DAX-I é um membro dos receptores nucleares conhecidos como receptores órfãos. Este gene demonstrou ser um poderoso repressor da transcrição de SFI e de vários genes. Os padrões de expressão de DAX-I são complementares daqueles de SFI, ambos expressos nas cristas genitais. Em resumo, dada a evidência exposta, desenvolveu-se a hipótese de que DAX-I é um antagonista de sry; esse antagonismo é dependente dos níveis relativos de DAX-I e sry e de um limiar que varia de espécie para espécie. A DAX-I foi classificada como o gene antitestículo. Na fêmea (cariótipo XX) é importante que ocorra a inativação de um dos cromossomas sexuais X para que se mantenha o equilíbrio genético ao igualar o conteúdo de DNA dos cromossomas. Esse cromossoma inativado constitui o chamado corpúsculo de Barr. No entanto, para que a meiose se realize, é necessário dos dois cromossomas X ativos nos ovócitos para assegurar a diferenciação ovárica e a fertilidade. 1.5 DIFERENCIAÇÃO DOS DUCTOS SEXUAIS O embrião possui, além das gônadas indiferenciadas, dois sistemas de ductos: os de potencialidade masculina denominam-se ductos de Wolff ou mesonéfricos, e os de potencialidade feminina se chamam ductos de Müller ou paramesonéfricos (figura 1). Se a diferenciação gonodal levou à formação de um testículo, a partir do ducto mesonéfrico ou de Wolff se desenvolverão os ductos eferentes, o epidídimo, os ductos deferentes e as vesículas seminais. As hormonas importantes no desenvolvimento do aparelho genital masculino são a testosterona, produzida pelas células de Leydig, e sua forma 5α reduzida, a 5α di-hidrotestosterona. Acredita-se que a testosterona é responsável pela virilização dos ductos de Wolff, e a di-hidrotestosterona dos órgãos genitais externos. No macho, os canais de Müller atrofiam-se devido à ação de uma hormona fetal de origem testicular denominada hormona inibidora das estruturas de Müller (HIM) ou hormona antimulleriana. Este processo começa assim que os cordões espermáticos se formam e se diferenciam as células de Sertoli. A existência de HIM foi proposta baseada em estudos realizados em bezerras freemartin, devido à existência de uma hormona responsável pela atresia dos ductos de Müller que na fêmea dá origem ao útero e aos ovidutos. Essa hormona provoca a involução do aparelho genital do bovino nas gestações gemelares nas quais os produtos de diferente sexo têm comunicação sanguínea por ter ocorrido a anastomose dos vasos de ambas as placentas (figura 3). A HIM é uma glicoproteína pertencente à subfamília de TGFβ, é expressa pelas células que darão origem às células de Sertoli e é um dos primeiros marcadores de diferenciação nestas células. A HIM é secretada na vida adulta pelas células de Sertoli no testículo e por células da granulosa no ovário. No rato a HIM é expressa-se no 12° dia em um teste padrão que segue muito de perto o aumento na expressão de sry. No macho, esta secreção de HIM continua durante a vida fetal e adulta, contudo os níveis de HIM declinam na puberdade devido a um aumento na secreção de testosterona. Vários fatores intervêm na regulação do gene de HIM, incluindo os acima descritos SFI e Sox9. O gene HIM contém segmentos de DNA que são conservados em várias espécies de vertebrados. Existe um nexo de ligação para SFI, que ativa a transcrição de HIM. A mutação no local de ligação de SFI resulta em reversão do sexo em indivíduos XY incluindo genitais femininos normais, presença de um útero formado enfatizando a importância de SFI na determinação sexual e na expressão de HIM. Embora SFI seja um bom candidato como regulador de HIM, é expresso em outras células, como as de Leydig e adrenais, que não expressam HIM. Em contrapartida, Sox9 é expresso unicamente nas células de Sertoli que são as produtoras de HIM. O gene HIM também tem um nexo de ligação para Sox9. Além disso, Sox9 pode atuar sinergicamente com SFI para promover a secreção de HIM. Ao contrário destes dois fatores de transcrição, DAX-I antagoniza a ação de Sox9 e provavelmente SFI sobre o promotor de HIM. Os ductos de Wolff tornam-se o sistema de ejaculação do macho. A porção mais próxima dos testículos dá origem ao epidídimo, a parte central ao ducto deferente e a porção mais distal às vesículas seminais. A próstata e a parte membranosa da uretra do macho desenvolvem-se a partir da porção pélvica do seio urogenital. A virilização e diferenciação dos ductos de Wolff dependem da produção de testosterona pelo testículo. Quanto aos órgãos genitais externos do macho, o tubérculo genital é ampliado e as dobras uretrais se fundem para formar a uretra peniana. A diferenciação dos órgãos genitais da fêmea ocorre de forma passiva, já que a ausência de testículos e por isso da hormona inibidora dos ductos de Müller (HIM), assim como dos andrógenos virilizantes, favorece o desenvolvimento dos ductos de Müller, enquanto os de Wolff sofrem atrofia. A porção cefálica dos ductos de Müller dá origem aos ovidutos, que na sua terminação caudal se fundem com o útero. O contato dos ductos de Müller com o seio urogenital induz uma intensa proliferação celular que resulta na formação da área uterovaginal localizado entre o seio urogenital e os ductos de Müller. As células do prato uterovaginal proliferam e aumentam a distância entre as duas estruturas criando o espaço que formará a vagina quando o prato é canalizado e forma um lúmen. Em contraste com o que ocorre no macho, na fêmea a maior parte do seio urogenital se mantém exposta na superfície da abertura onde desembocam a vagina e a uretra. O tubérculo urogenital da fêmea tem um crescimento limitado e forma o clitóris. A sequência de passos da diferenciação sexual do aparelho genital é resumida na tabela 1. Assim, da mesma maneira que a gônada e os ductos sexuais se desenvolvem para o tipo feminino ou masculino, propôs-se que o cérebro pode ser "masculinizado" ou permanecer "Feminizado". A diferenciação do hipotálamo vai depender do ambiente esteroidal do neonato e ocorre na fase perinatal. Estes eventos serão de grande transcendência na vida reprodutiva do indivíduo. Tanto a fêmea como o macho nascem com a capacidade de secreção de gonadotropinas de acordo com um padrão cíclico; contudo, no macho, a exposição do hipotálamo à ação dos andrógenos testiculares durante os primeiros dias da vida extrauterina provoca a masculinização, com o qual o hipotálamo do macho é programado para que a secreção de gonadotropinas se realize a um ritmo relativamente constante por parte da hipófise (secreção tônica). Na fêmea, tanto a secreção tônica como a cíclica se conservam. No entanto, observou-se que a injeção de testosterona ou o transplante de testículo na rata fêmea durante os primeiros dias de vida, suprime a sua futura atividade estral (secreção cíclica). Por outro lado, se os ovários forem transplantados para o rato macho normal castrado na idade adulta, o animal não desenvolve qualquer atividade cíclica, mas se os machos transplantados forem castrados ao nascer, o ovário é capaz de efetuar mudanças cíclicas e ovulações. Isto foi demonstrado em roedores, mas não em animais domésticos ou na espécie humana. Portanto, o padrão de secreção de gonadotropinas, seja cíclico ou tônico, não depende da hipófise, mas do hipotálamo e sua correta diferenciação. 1.7 CONCLUSÕES A maioria dos conhecimentos no campo da biologia do desenvolvimento e, muito especificamente, dos processos de diferenciação sexual têm sido originados de estudos relacionados com desordens congênitas, que na sua maioria devem-se a defeitos de genes específicos. A análise detalhada destas desordens permitiu entender alguns mecanismos endócrinos, moleculares e genéticos envolvidos na diferenciação sexual. A identificação do gene sry como determinante do testículo foi uma contribuição crucial e abriu as portas à compreensão dos mecanismos moleculares e celulares relacionados com o desenvolvimento do testículo. Se este gene não estiver presente, é criado um programa genético alternativo para levar a cabo a diferenciação gonodal para o ovário. In this paper I argue that Miranda Fricker’s account of blame in “What’s the Point of Blame? A Paradigm Based Explanation” can assist in explaining why cancel culture is ultimately unproductive. In particular, the phenomenon of cancel culture possesses pathological forms of blame. There are three specific pathologies outlined by Fricker that can be observed in cancel culture. They are as follows: cancel culture does not leave room for people to learn from their mistakes, it does not express its blame (...) in the proper ethical register, and cancel culture allows for blame to fester and spread. In the first half of my paper, I will lay out the distinct aspects of Fricker’s paper that relate to cancel culture and a definition of the term cancel culture. In the second half, I will explore the real-life cancelation of actor Lea Michele so as to validate the presence of cancel culture in our society today. Furthermore, I will expand on three of Fricker’s pathologies that are present in cancel culture and refute a counter argument people may pose who are supportive of cancel culture. (shrink)

The goal of this article is to compare and contrast the traditional Western versus the postmodern colonization of the mind. How is the current technological age barbaric? I investigate Aimé Césaire’s writings, refer to Lea Ypi’s definition of colonialism, and discuss society’s use of psychopolitics to find the answer.

Why should musical compositions and artistic images be included among the types of expression covered by free speech principles? One way to answer this question is to show how expression in nonverbal media can be functionally similar to other types of verbal expression. But this leaves us with an intuitively unsatisfying explanation of why free speech principles cover nonverbal creative expression that does not functionally emulate literal speech. In this article, as an alternative justification, we develop and defend the idea (...) that musical and pictorial expression‚ much like literal speech, can be media through which people think aloud, as opposed to mere tools for the transmission of thought. We use this proposal to provide a more robust justification for including nonverbal creative expression in the scope of free speech coverage, and we outline some of the practical policy implications that come with adopting this justificatory strategy. A surprising feature of the White Rose anti-Nazi resistance pamphlets is their appeal to a foundational classical Chinese text, the Laozi (otherwise known as the Daodejing), to buttress their critique of fascism and authoritarianism. I argue that from the perspective of a 1942 educated readership, the act of quoting the Laozi functioned as a subtle and pointed nod to anti-fascist intellectuals in pre-war Germany, many of whom had interpreted the Laozi as an anti-authoritarian and pacifist text. To a sympathetic reader, (...) the Laozi therefore constituted an apt reference point for critiquing National Socialism. I then introduce a complication for this wartime reading of the Laozi from the perspective of its ambiguous reception in ancient Chinese political thought. I more specifically discuss an ancient line of interpretation of the Laozi that points in the direction of authoritarianism — in stark tension with the White Rose message of passive resistance and popular revolt. Reflection on the historical injustice suffered by many formerly colonized groups has left us with a peculiar account of their claims to material objects. One important upshot of that account, relevant to present day justice, is that many people seem to think that members of indigenous groups have special claims to the use of particular external objects by virtue of their attachment to them. In the first part of this paper I argue against that attachment-based claim. In the second part (...) I suggest that, to provide a normatively defensible account of why sometimes agents who are attached to certain external objects might also have special claims over them, the most important consideration is whether the agents making such claims suffer from structural injustice in the present. In the third part I try to explain why structural injustice matters, in what way attachment-based claims relate to it and when they count. (shrink)

A well-known debate in normative ethics is that between proponents of Act Consequentialism and Rule Consequentialism. Given the structural similarities between Rule Consequentialism and existing forms of Contractualism, one might expect a similar debate to arise among contractualists. However, this is not the case. Some, following T. M. Scanlon, even argue that this question is “misconceived” – that there is something deeply mistaken about considering the possibility of an act-based form of contractualism. In this paper, I challenge this claim.

Most current dental ethics curricula use a deontological approach to biomedical and dental ethics that emphasizes adherence to duties and principles as properties that determine whether an act is ethical. But the actual ethical orientation of students is typically unknown. The purpose of the current study was to determine the ethical orientation of dental students in resolving clinical ethical dilemmas. First-year students from one school were invited to participate in an electronic survey that included eight vignettes featuring ethical conflicts common (...) to the health care setting. The Multidimensional Ethics Scale was used to evaluate the students’ ethical judgments of these conflicts. Students rated each vignette along 13 ethically relevant items using a 7-point scale. Nine of the thirteen items were analyzed because they represent the dominant ethical theories, including deontology. One hundred sixteen dental students successfully completed the survey. Of the analyzed items, those associated with deontology had comparatively weak associations with whether students judged the action to be ethical and whether students judged themselves likely to perform the action. Whether an action was judged to be caring had the strongest association with whether the action was judged to be ethical and whether students judged themselves likely to perform the action. These results suggest that adherence to duties or principles has weaker association with students’ ethical judgments and behavior compared to caring, which was found to be more influential in their ethical judgments and behavior. Abstract: Dates are the fruit of date palm trees, and it is one of the fruits famous for its high nutritional value. It is a summer fruit spread in the Arab world. In the past, the Arabs relied on it in their daily lives. Dates take an oval shape and vary in size from 20 to 60 mm in length and 8 to 30 mm in diameter. The ripe fruit consists of a hard core surrounded by a papery cover called (...) the tartar that separates the core from the fleshy part that is eaten. Historians disagreed about the place of its origin, so some historians expressed their belief that it had originated around the Arabian Gulf, and some of them say that the oldest known information about palm trees was in Babylon 4 thousand years ago BC. It is known about the ancient Egyptians using dates in wine. Dates contain a high nutritional value and are considered a basic food for man since ancient times. The fruits of dates are considered the highest fruits in containing sugars. These components vary according to the nature of the fruit, whether it is wet, semi-dry or dry, as well as according to the environmental conditions surrounding the trees. The components of the fruits also differ in different varieties and increase the percentage of sugars in dates is 70-78% of the fruit’s components. These sugars are characterized by their rapid absorption and transfer into the blood directly, digestion and burning. 10 dates (about 100 grams) per day provide a person with all his daily needs of magnesium, manganese, copper, sulfur and half of his needs of calcium and potassium. In this paper we presented a system that recognize the nine types of dates based on deep learning using python on Colab, and classifying using a dataset contain 1350 images. Our trained model achieved an accuracy of 99.44% on a proven test set. (shrink)

Abstract: The Knowledge-Based System for Diagnosing Breast Cancer aims to support medical students in enhancing their education regarding diagnosis and counseling. The system facilitates the analysis of biopsy images under a microscope, determination of tumor type, selection of appropriate treatment methods, and identification of disease-related questions. According to the Ministry of Health's annual report in Gaza, there were 7,069 cases of breast cancer between 2009 and 2014, with 1,502 cases reported in 2014. In an era dominated by visual information, where (...) 65% of the population are visual learners, the Knowledge-Based System serves as an effective tool to aid long-term information retention by utilizing visual and textual resources. The system features a user-friendly interface, enabling students to diagnose the disease and expand their understanding. Furthermore, it can be accessed via smartphones. The Knowledge-Based System comprises three primary components: Firstly, it provides specific patient-oriented questions to familiarize students with the questioning process. Secondly, it facilitates the analysis of biopsy sample images to determine their integrity and identify potential infections. Abstract: Colon cancer is a prevalent and life-threatening disease, necessitating accurate and timely diagnosis for effective treatment and improved patient outcomes. This research paper presents the development of a knowledge-based system for diagnosing colon cancer using the CLIPS language. Knowledge-based systems offer the potential to assist healthcare professionals in making informed diagnoses by leveraging expert knowledge and reasoning mechanisms. The methodology involves acquiring and structuring medical knowledge specific to colon cancer, followed by the implementation of a knowledge- based system using (...) CLIPS. The system's design integrates relevant medical databases and employs advanced reasoning mechanisms to facilitate accurate diagnosis. Evaluation results demonstrate the system's effectiveness, with high accuracy and sensitivity rates compared to existing methods. The findings highlight the potential of knowledge-based systems in improving clinical practice and patient care. The research contributes to the growing field of medical informatics by demonstrating the feasibility and effectiveness of CLIPS-based knowledge systems for colon cancer diagnosis. The problem of other minds has a distinguished philosophical history stretching back more than two hundred years. Taken at face value, it is an epistemological question: it concerns how we can have knowledge of, or at least justified belief in, the existence of minds other than our own. In recent decades, philosophers, psychologists, neuroscientists, anthropologists and primatologists have debated a related question: how we actually go about attributing mental states to others (regardless of whether we ever achieve knowledge or rational (...) justification in this domain). Until the mid-nineties, the latter debate – which sometimes goes under the name of the “mindreading” debate – was characterized by a fairly clear-cut opposition between two theoretical outlooks: “theory-theory” (TT) and “simulation theory” (ST). Theory-theorists typically argued that we attribute mental states to others on the basis of a “theory of mind” that is either constructed in early infancy and subsequently revised and modified (Gopnik 1996), or else is the result of maturation of innate mindreading “modules” (Baron-Cohen 1995). Abstract: Ensuring the proactive detection of transaction risks is paramount for financial institutions, particularly in the context of managing credit scores. In this study, we compare different machine learning algorithms to effectively and efficiently. The algorithms used in this study were: MLogisticRegressionCV, ExtraTreeClassifier,LGBMClassifier,AdaBoostClassifier, GradientBoostingClassifier,Perceptron,RandomForestClassifier,KNeighborsClassifier,BaggingClassifier, DecisionTreeClassifier, CalibratedClassifierCV, LabelPropagation, Deep Learning. The dataset was collected from Kaggle depository. It consists of 164 rows and 8 columns. The best classifier with unbalanced dataset was the LogisticRegressionCV. The Accuracy 100.0%, precession 100.0%,Recall100.0% and the F1-score (...) 100.0%. However, the best classifier with balanced dataset was the LogisticRegressionCV. They are a good source of protein, vitamins, and minerals and play a significant role in human diets, especially in coastal and island communities. In addition, fishing and fish farming are major industries that provide employment and income for millions of people worldwide. Moreover, fish play a critical role in marine ecosystems, serving as prey for larger predators and helping to maintain the balance of aquatic food chains. Overall, fish play (...) a vital role in supporting human well-being and the health of our planet. . Fishes have a lots of types such that: Red Mullet, Sea Bass, Striped Red Mullet and Shrimp. Each of them has its own shape and characteristics that differ from other types. We proposed a system that recognize nine types of fishes using deep learning. We trained the model with a dataset that contain 9000 images that were slit into 6300 images for training, 1350 for validation and 1350 for testing. The proposed model achieved accuracy (99.68%), precision (99.69%), recall (99.68%), and f1-score (99.68%). Risk Management Strategies and Investment Behaviors are considered important factors in the investing activities of the retail investors. This study seeks to determine the relationship between Risk Management Strategies and Investment Behavior of Generation Z retail investors. The study is a correlational research and purposive sampling was used to select the respondents for this study. Cochran’s formula was utilized to determine the total sample size or total number of respondents. Spearman’s Rank-Order Correlation was employed to assess the significant relationship of (...) Risk Management Strategies and Investment Behaviors. Based on the results, it showed that risk management strategies have a significant relationship with one or more of the investment behaviors. Risk avoidance has a significant relationship with confirmation bias. Risk reduction has a significant relationship with all of the investment behaviors such as overconfidence, loss-aversion, recency bias, and confirmation bias. Risk transfer shows a significant relationship with only loss-aversion and confirmation bias. And lastly, risk acceptance also has a significant relationship with all of the investment behaviors. This study recommends exploring other types of behavioral biases under cognitive bias and emotional bias to determine their correlation with risk management strategies. Also, broadening the sample size, demographic scope, and geographic scope of the study can improve the generalizability of the study and provide a more comprehensive understanding of the relationship between investment behavior and risk management strategies. The researchers also recommend to utilize interviews and focus group discussions to gain an in-depth understanding of the underlying values and attitudes concerning investment behavior. (shrink)

Academic excellence and intelligence are commonly lauded as commendable attributes synonymous with success. However, a disconcerting trend has surfaced within educational institutions, challenging the prevailing narrative of scholastic accomplishment—smart shaming. This research delves into the increasing concern of smart shaming within educational settings, particularly at Immaculate Conception College of Balayan, Inc., questioning the predominant emphasis on academic excellence and intelligence. A qualitative case study design, along with judgmental sampling, was employed to examine fifteen (15) student leaders who had experienced smart (...) shaming on campus. Thematic analysis revealed that sarcastic remarks including "edi wow," "Okay ikaw na matalino," "bidabida," and "sipsip" were a prevalent manifestation of smart shaming, adversely affecting emotional well-being, academic and leadership prowess, mental health, and self-perception. Coping mechanisms, predominantly centered around ignoring and focusing on goals, were identified. The findings underscore the imperative for educators to remain vigilant regarding instances of smart shaming in classrooms, promoting open communication to ensure student well-being. Additionally, guidance counselors play a pivotal role in aiding students in effectively navigating the repercussions of smart shaming. This study advocates for proactive programs and interventions to address and alleviate the deleterious effects of smart shaming on students. (shrink)

We propose a formal framework for interpretable machine learning. Combining elements from statistical learning, causal interventionism, and decision theory, we design an idealised explanation game in which players collaborate to find the best explanation for a given algorithmic prediction. Through an iterative procedure of questions and answers, the players establish a three-dimensional Pareto frontier that describes the optimal trade-offs between explanatory accuracy, simplicity, and relevance. Multiple rounds are played at different levels of abstraction, allowing the players to explore overlapping causal (...) patterns of variable granularity and scope. We characterise the conditions under which such a game is almost surely guaranteed to converge on a optimal explanation surface in polynomial time, and highlight obstacles that will tend to prevent the players from advancing beyond certain explanatory thresholds. The game serves a descriptive and a normative function, establishing a conceptual space in which to analyse and compare existing proposals, as well as design new and improved solutions. (shrink)

Abstract: People all over the world have been drinking tea for thousands of centuries, and for good reason. Many types of teas can help you stay healthy by boosting your immune system, reducing inflammation, and even preventing cancer and heart disease. There is sufficient material to show that regularly consuming tea can improve your health over the long term. A deep learning model that categorizes tea disorders has been completed. When focusing on the tea, we must also focus on and (...) take into account the diseases that harm it. The aim of this paper is to diagnose tea diseases based on a dataset containing 8000 picture and 8 classes .The model gave the final accuracy test with an F1-score of 99.41% in an estimated time 3 second, as with these results it proved its effectiveness and good ability in the classification test. Abstract. Intelligent Tutoring System Builder (ITSB) is an authoring tool designed and developed to aid teachers in constructing intelligent tutoring systems in a multidisciplinary fields. The teacher is needed to create a set of pedagogical fundamentals, which, in line, are inured to automatically build up a broad tutor framework and construct an intelligent tutoring system. In this paper an explanation of the theory and the architecture of the tool is outlined. A presentation of several system components, the requirements of the (...) different components, integration of these components in ITSB tool are shown. Furthermore, implanting of requirements, cognitive principle, and common design fundamentals in the tool to ease the use of teachers. Almost everyone who has a limb amputated will experience a phantom limb--the vivid impression that the limb is not only still present, but in some cases, painful. There is now a wealth of empirical evidence demonstrating changes in cortical topography in primates following deafferentation or amputation, and this review will attempt to relate these in a systematic way to the clinical phenomenology of phantom limbs. With the advent of non-invasive imaging techniques such as MEG (magnetoencephalogram) and functional MRI, topographical reorganization (...) can also be demonstrated in humans, so that it is now possible to track perceptual changes and changes in cortical topography in individual patients. We suggest, therefore, that these patients provide a valuable opportunity not only for exploring neural plasticity in the adult human brain but also for understanding the relationship between the activity of sensory neurons and conscious experience. We conclude with a theory of phantom limbs, some striking demonstrations of phantoms induced in normal subjects, and some remarks about the relevance of these phenomena to the question of how the brain constructs a 'body image.'. There is a loud and persistent drum beat of support for schools, for citizenship, for diversity and inclusion, and increasingly for labor market readiness with very little critical attention to the assumptions underlying these agendas, let alone to their many internal contradictions. Accordingly, in this book I examine the philosophical, motivational, and practical challenges of education theory, policy, and practice in the twenty-first century. As I proceed, I do not neglect the historical, comparative international context so essential to better understanding (...) where we are, as well as what is attainable in terms of educational justice. Aristotle’s dialogue Protrepticus is not only his earliest work of ethics but also the root of all his subsequent investigations into ethics. Here we explore the various ways Aristotle retained in memory the contents of the Protrepticus and redeployed them in the Eudemian Ethics, including the common books. Since Aristotle himself does not explicitly acknowledge the foundational significance of the Protrepticus to his later works, our exploration must proceed on the basis of our knowledge of the earlier work, which can (...) then related to comparable passages in the treatises. But the Protrepticus is a lost work, unlike the EE and the EN, in the sense that no medieval manuscripts of it have survived. And so it first needs to be reconstructed on the basis of the streams of reception that deposited information in surviving works by later ancient authors. We begin in part 1 by reviewing the surviving evidence of the Protrepticus before presenting our wider reconstruction and conception of the work as a dialogue in part 2. In part 3, we discuss ideas and arguments in the reconstructed dialogue that are retained and redeployed in the Eudemian Ethics, focussing on the account of intellectual virtues in EE v and the introduction and methodological framework in EE i-ii.1. In this paper I identify and respond to four persistent objections to the idea of wilderness: empirical, cultural, philosophical and environmental. Despite having dogged the wilderness debate for decades, none of these objections withstands scrutiny; rather they are misplaced criticisms that hinder fruitful discussion of the philosophical ramifications of wilderness by needlessly complicating the idea itself. While there may be other justifiable concerns about the idea of wilderness, it is time to move beyond the four discussed in this paper.

Persons think. Bodies, time-slices of persons, and brains might also think. They have the necessary neural equipment. Thus, there seems to be more than one thinker in your chair. Critics assert that this is too many thinkers and that we should reject ontologies that allow more than one thinker in your chair. I argue that cases of multiple thinkers are innocuous and that there is not too much thinking. Rather, the thinking shared between, for example, persons and their bodies is (...) exactly what we should expect at the intersection of part sharing and the supervenience of the mental on the physical. In this paper we show how a student can learn the basics of the system databases using (W3school CSS) which was built as intelligent tutoring educational system by using the authoring tool called (ITSB). The learning material contains CSS and HTML. We divided the material in a group of lessons for novice learner which combines relational system and lessons in the process of learning. The student can learn using example of CSS, and types of CSS color. Furthermore, the intelligent tutoring (...) system supports not only lessons; but exercises of different difficult levels for each lesson. When a student finish successfully the first difficulty level in a lesson, the student is allowed to move to the next difficulty level of the exercises of the lesson. (shrink)

The research aims to identify the trends of Palestinian higher educational institutions in Gaza Strip as learning organizations from the perspective of senior management in the Palestinian universities in Gaza Strip. The researchers used descriptive analytical approach and used the questionnaire as a tool for information gathering. The questionnaires were distributed to senior management in the Palestinian universities. The study population reached (344) employees in senior management is dispersed over (3) Palestinian universities. A stratified random sample of (182) employees from (...) the Palestinian universities was selected and the recovery rate was (69.2%). Statistical analysis (SPSS) program was used for analysis and processing the data. The study found the following results: There is an agreement about: the importance of the focus of "organizational structure" with an average approval, the importance of "technological infrastructure" axis with high approval, and the importance of "strategic" with an average approval. The results concluded that the study sample agree on the importance of "organizational dimension" highly. The results showed that the sample believe that the "strategic leadership" level in the universities got medium-approval. There is a fair level agreement about the axis of interest "teams / committees". There is an agreement about the importance of the "human dimension" moderately. The results showed that the sample is highly agreed about the importance of focus of the "knowledge management", and the focus of the "continuing education". They agreed weakly to somewhat about the importance of the focus of "scientific research", moderately agree about the importance of the center of "institutional culture", agree moderately on the importance of "cognitive dimension". The results showed that the sample largely agree on the importance of the focus of "strategic partnerships and alliances". They agree highly on the axis "keep up with the labor market" and on the focus of the importance of "technology incubators". The results showed that the sample moderately agree about the importance of the theme on "consulting and training", the importance of the focus of "social responsibility", and the focus of the "Community dimension". The study found a group of recommendations including: there is a need to provide suitable environment that achieve learning organizations. There is a need to develop the technological infrastructure (hardware, software, networks, databases, and human skills) because of the great advantages that they offer. The universities need to adopt knowledge management in the academic and administrative departments because knowledge is the core of the work of these departments. The establishment of technology incubators in universities to adopt outstanding university research projects, to protect, to supports, and to market them; furthermore, to develop the capabilities and skills of employees in the field of information technology. The study determined the chemical composition and acceptability of organic bath soap among students in Yaba College of Technology, Yaba, Lagos State, Nigeria. The study was guided by two research questions. Mixed method research design was used for the study. Population was 46 which comprised all Home Economics students from the Department of Vocational Education. In-home use questionnaire was used for data collection. The study produced two samples of turmeric bath soap. Findings showed that the chemical compositions of the turmeric (...) soap samples are in accordance with the specifications of the Standard Organization of Nigeria (SON) with toilet soap specifications of pH ranging from 9 to 11, total fatty matter ranging from 60-75 gm and total free caustic soda ranging from 0.05 to 0.08 %. Findings showed that the sensory evaluation attributes for colour, smell, consistency foam ability and skin sensitivity were accepted after use for three months as the samples were skin friendly, safe to use without side effects. It was recommended that turmeric body soap and turmeric face gel can be produced in large quantities for commercial purpose as a form of income generation for unemployed graduates. (shrink)

The study aimed to investigate the reality of spreading the culture of entrepreneurship at Al-Azhar University from the point of view of students of the Faculty of Engineering and Information Technology and diagnose the most important obstacles that limit its activation. The researchers used the descriptive approach (survey) to achieve the objectives of the study, and relied on the questionnaire as a tool for applied study. The study concluded that: The reality of spreading the culture of entrepreneurship at Al-Azhar University (...) from the point of view of the students participating in the study came with an average degree of approval, while the most important obstacles and possible proposals for activating it came with a high degree of approval, which indicates the existence of strengths of Al-Azhar University in spreading the culture of entrepreneurship among students. Deficiencies must be corrected through proposals. In light of the results of the study, it was possible to develop a number of recommendations that can contribute to activating the culture of entrepreneurship at Al-Azhar University. Including the establishment of a specialized center for entrepreneurship within the university, develop a declared strategic plan to spread and develop the culture of entrepreneurship, allocate a sufficient budget to sponsor and support entrepreneurial ideas and projects for students and provide specialized cadres and certified trainers in the field of entrepreneurship. This comparative examination of Nietzsche and the Islamic philosopher al-Kindi emphasizes their mutual commitment to the recovery of classical Greek and Hellenistic thought and the idea of philosophy as a way of life. Affiliating both thinkers with the Stoic lineage in particular, I examine the ways in which they appropriate common themes such as fatalism, self-cultivation via spiritual exercises, and the banishing of sorrow. Focusing primarily on their respective conceptions of self and nature, I argue that the antipodal worldviews of (...) al-Kindi and Nietzsche can be understood as a bifurcation of Stoic philosophy. In this paper, I offer an expansionist view of the Frickerian central case of testimonial injustice, citing examples from the South Asian context. To defend this expansionist position, I provide an argument in three parts. First, I argue that credibility deficit and credibility excess are entangled with each other in such a way that often, one produces the other. Secondly, I contend that we should not say that systematic testimonial injustice is a consequence of credibility deficit only because of the (...) entanglement between them. I also contend that for being the central case of testimonial injustice, identity prejudice should not be necessarily negative; it can be positive as well. Propounding a twofold condition of the status of a knower, the last part claims that testimonial injustice occurs when one of the two conditions remains unmet. (shrink)
